摘要
旨在了解河南地区猪繁殖与呼吸综合征病毒(PRRSV)的流行现状及变异情况。对在河南省信阳地区某疑似PRRSV阳性猪场获得的肺脏组织进行病毒检测和分离,结果在MARC-145细胞上成功分离到1株PRRSV,命名为HNxy23。对HNxy23序列分析显示,HNxy23基因组全长15049bp,与美洲型经典毒株VR-2332同源性为84.1%,与其他美洲型毒株JXA1、NADC30、CHsk1401和15LN1的同源性分别为83.6%、90.2%、90.2%和91.0%。Nsp2氨基酸序列比对结果显示,HNxy23与NADC30毒株相似存在131(111+1+19)个不连续的氨基酸位点缺失。GP5氨基酸序列比对结果显示,HNxy23的抗原表位存在部分氨基酸突变。经重组分析表明,HNxy23是谱系1的NADC30毒株与谱系8的高致病性类疫苗毒株JXA1-P80的重组毒株,重组区域为5392~6969bp。本研究结果丰富了河南地区PRRSV分子流行病学数据,也警示当前PRRSV的遗传变异正在加剧。
Abstract
To investigate the epidemiological status and genetic variation of porcine reproductive and respiratory syndrome virus (PRRSV) in Henan Province, lung tissue samples obtained from a suspected PRRSV-positive pig farm in Xinyang were tested and subjected to virus isolation. PRRSV was successfully isolated on MARC-145 cells and designated as HNxy23. Subsequent sequence analysis of HNxy23 revealed the following facts: The full length of the HNxy23 genome was 15049 bp. It possessed a homology of 84.1% with the classic American strain VR-2332, and had homologies of 83.6%, 90.2%, 90.2%, and 91.0% with JXA1, NADC30, CHsk1401, and 15LN1, respectively. The amino acid sequence alignment of Nsp2 showed that HNxy23, similar to the NADC30 strain, had a total of 131 discontinuous amino acid site deletions, including 111+1+19. GP5 amino acid sequence alignment revealed partial amino acid mutations in the antigenic epitopes of HNxy23. The recombination analysis confirmed HNxy23 to be a recombinant strain between lineage 1 NADC30 and lineage 8 highly pathogenic vaccine strain JXA1-P80, with a recombination region of 5392 bp to 6969 bp.
