摘要

目的 研究泛素特异性蛋白酶7(USP7)对人结肠癌HCT116细胞干性特征及奥沙利铂(OXA)耐药的影响。方法 构建HCT116 OXA耐药细胞株,检测USP7的表达水平;分别转染siCON、USP7-si1和USP7-si2,采用免疫印记法验证敲减效率;通过克隆形成实验检测细胞增殖能力;qPCR实验检测结肠癌细胞干性相关基因的mRNA水平;CCK-8实验检测细胞对OXA的敏感性。结果 USP7在OXA耐药HCT116细胞中表达水平上调,敲减USP7能够显著抑制HCT116细胞的增殖能力(P<0.01),抑制结肠癌干性相关基因的表达和OXA耐药性(P<0.01)。结论 USP7能够促进结肠癌HCT116细胞的干性特征并介导OXA耐药。

Abstract

Objective To investigate the effects of USP7 on the stemness characteristics and oxaliplatin resistance of human colon cancer HCT116 cells. Methods Oxaliplatin-resistant HCT116 cell line was established, and the expression level of USP7 was detected. Si-CON, USP7-si1, and USP7-si2 were transfected respectively, and the knockdown efficiency was verified by immunoblotting. Colony formation assays were used to assess cell proliferation ability. qPCR was performed to detect the expression levels of cancer stemness-related genes. CCK-8 assays were employed to evaluate the sensitivity of cells to oxaliplatin. Results USP7 expression was upregulated in oxaliplatin-resistant HCT116 cells. Knockdown of USP7 significantly inhibited the proliferation ability of HCT116 cells (P<0.01) and suppressed the expression of cancer stemness-related genes and oxaliplatin resistance (P<0.01). Conclusion USP7 can promote the stemness characteristics of colon cancer HCT116 cells and mediate oxaliplatin resistance.
