五苓散通过NF-κB/NLRP3信号通路抑制糖尿病肾病小鼠肾纤维化

五苓散通过NF-κB/NLRP3信号通路抑制糖尿病肾病小鼠肾纤维化
Wuling Powder Inhibits Renal Fibrosis in Diabetic Nephropathy Mice via NF-κB/NLRP3 Signaling Pathway
目的 探究五苓散对糖尿病肾病db/db小鼠肾纤维化的改善作用及机制。 方法 将24只8周龄db/db小鼠按照体质量随机分成模型组、五苓散组、厄贝沙坦组,每组8只,另取8只同周龄db/m小鼠为正常组。五苓散组给予五苓散7.2 g/(kg·d)灌胃,厄贝沙坦组给予厄贝沙坦22.5 mg/(kg·d),正常组和模型组db/db小鼠给予等体积生理盐水,连续给药8周。观察4组小鼠一般情况,检测糖化血清蛋白(GSP)、24 h尿蛋白定量(24 h-UTP)、甘油三酯(TG)、总胆固醇(TC)、血肌酐(SCr)及尿素氮(BUN)含量;Masson染色观察肾组织病理改变;Western blot、qPCR法检测肾组织中核因子NF-κB p65、核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)mRNA表达水平和蛋白表达量。 结果 与正常组比较,模型组db/db小鼠体质量及GSP、24 h-UTP、TG、TC、SCr、BUN含量均明显升高(P<0.05);肾小球体积增大,基底膜增厚,肾间质大量胶原纤维沉积;肾组织中NF-κB p65、NLRP3 mRNA表达水平及蛋白表达量均明显升高(P<0.05);与模型组比较,五苓散组db/db小鼠治疗后体质量及GSP、24 h-UTP、TG、TC、SCr、BUN含量均明显降低(P<0.05),厄贝沙坦组db/db小鼠24 h-UTP、SCr、BUN含量均明显降低(P<0.05);五苓散组和厄贝沙坦组db/db小鼠肾小球基底膜增厚明显改善,肾间质胶原纤维沉积明显减少,肾组织中NLRP3、NF-κB p65 mRNA表达水平及蛋白表达量均明显降低(P<0.05)。 结论 五苓散可能通过调控NF-κB/NLRP3信号通路,抑制糖尿病肾病db/db小鼠肾脏组织纤维化。
Objective To investigate the effect and mechanism of Wuling Powder on renal fibrosis in db/db mice with diabetic nephropathy. Methods According to body weight, 24 8-week-old db/db mice were randomly divided into model group, Wuling Powder group and irbesartan group, 8 db/db mice in each group, and 8 db/m mice of the same age were selected as normal group. The Wuling Powder group was given Wuling Powder 7.2 g/(kg·d) by gavage, the irbesartan group was given irbesartan 22.5 mg/(kg·d), db/db mice in the normal group and model group were given equal volume normal saline, all groups were intervened for 8 weeks. The general situation of mice were observed, the content of glycated serum protein (GSP), 24 hour urine total protein quantity (24 h-UTP), triglyceride (TG), total cholesterol (TC), serum creatinine (SCr), and blood urea nitrogen (BUN) were tested; the pathological changes of renal tissue were observed by Masson staining; Western blot and qPCR were used to detect the mRNA and protein expression of nuclear factor NF-κB p65 and NOD-like receptor protein 3 (NLRP3) in renal tissue. Results compared with the normal group, the body weight, the content of GSP, 24 h-UTP, TG, TC, SCr, and BUN of db/db mice in the model group increased significantly (P<0.05); the volume of glomerulus increased, the glomerular basement membrane thickened, and a large amount of collagen fibers deposited in renal interstitium; the mRNA and protein expression of NF-κB p65 and NLRP3 in renal tissues increased significantly (P<0.05). Compared with the model group, the body weight, the content of GSP, 24 h-UTP, TG, TC, SCr, and BUN of db/db mice in the Wuling Power group decreased significantly (P<0.05) after treatment, the content of 24 h-UTP, SCr and BUN of db/db mice in the irbesartan group significantly decreased (P<0.05); the glomerular basement membrane thickening of db/db mice in the Wuling Powder and irbesartan groups was significantly improved, and the collagen fibers deposition in renal interstitium reduced significantly, the mRNA and protein expression of NF-κB p65 and NLRP3 in renal tissues significantly decreased (P<0.05). Conclusion Wuling Powder may inhibit renal fibrosis in db/db mice with diabetic nephropathy by regulating NF-κB/NLRP3 signaling pathway.
