一株野生白耙齿菌生物学特性及其降解柠条的效果研究

一株野生白耙齿菌生物学特性及其降解柠条的效果研究
Biological Characteristics of a Wild Irpex lacteus Strain and Its Effect on Degrading Caragana korshinskii
针对沙区柠条平茬资源饲料化进程慢的现实问题,在柠条枯死枝上分离得到一株野生白耙齿菌n6,对该菌株的生物学特性及其降解柠条后对柠条营养和木质纤维素的效果进行初步研究。采用单因素和正交试验探究该菌的生物学特性,同时测试该菌发酵对柠条营养变化和木质纤维素降解能力的影响。单因素试验结果表明,白耙齿菌最适碳源为麦芽糖,最适氮源为谷氨酸,温度适宜范围为25~35 ℃,pH值最适范围为4.5~6.0。经过正交试验筛选适宜的培养基配方为:麦芽糖20 g,蛋白质2 g,pH值4.5,温度30 ℃,琼脂20 g和水1 L。扩繁菌种筛选最适宜培养基配方为:荞麦98%,葡萄糖1%,石膏1%。用获得的白耙齿菌n6进行柠条揉丝碎屑的发酵,结果表明:大部分柠条常规营养有显著提高,其中饲料的重要指标粗蛋白含量显著高于原样;中性洗涤纤维、酸性洗涤纤维和酸不溶木质素含量均比原样组显著降低,降解率分别达到8.10%、10.46%和18.67%,说明利用白耙齿菌n6降解柠条木质纤维素具有较好的应用潜力。
To address the slow progress in the forage utilization of pruned Caragana korshinskii in sandy areas, a wild Irpex lacteus strain n6 was isolated from withered branches of C. korshinskii. The biological characteristics of this strain and its effects on the nutrient composition and lignocellulose of C. korshinskii after degradation were preliminarily studied. Single-factor and orthogonal experiments were conducted to investigate the biological characteristics, and the effects of fermentation with this strain on nutrient changes and lignocellulose degradation capacity of C. korshinskii were tested. The single-factor experiment results show that the optimal carbon source for I. lacteus is maltose and the optimal nitrogen source is glutamic acid. The suitable temperature range is 25-35 °C, and the optimal pH range is 4.5-6.0. The orthogonal experiment screens the suitable culture medium formula as follows: 20 g of maltose, 2 g of peptone, pH 4.5, temperature of 30 °C, 20 g of agar, and 1 L of water. The optimal culture medium formula for strain propagation was buckwheat 98%, glucose 1%, and gypsum 1%. The fermentation of rubbed and chopped C. korshinskii using the obtained I. lacteus n6 shows that most conventional nutrients in C. korshinskii are significantly increased, with crude protein content, an important feed indicator, being significantly higher than that in the original sample. The contents of neutral detergent fiber, acid detergent fiber, and acid-insoluble lignin are all significantly reduced compared with that of the original sample, with degradation rates reaching 8.10%, 10.46%, and 18.67%, respectively. These results indicate that the use of I. lacteus n6 for lignocellulose degradation of C. korshinskii has good application potential.
