盐酸提取大眼金枪鱼骨中可溶性钙工艺优化
Optimization of soluble calcium extraction process from bigeye tuna (Thunnus obesus) bones using hydrochloric acid
目的 充分提取鱼骨钙,高值化利用鱼骨资源。 方法 以大眼金枪鱼骨为原料,采用盐酸提取法提取可溶性钙。大眼金枪鱼骨中钙含量采用高效液相色谱-电感耦合等离子体质谱(HPLC-ICPMS)测定,提取液中的可溶性钙采用原子吸收光谱法测定.在单因素试验优化盐酸浓度、液料比、提取温度和提取时间的基础上,采用Box-Behnken设计对盐酸浓度、液料比和提取时间三个因素进一步优化。进行模型可行性分析,将钙提取率的实际值和多元回归拟合得到的预测值进行比对。 结果 大眼金枪鱼骨中钙含量高达164.7 mg/g。通过Box-Behnken设计拟合的方程可用于实际情况的模拟,修正后工艺参数为盐酸浓度1.00 mol/L、液料比14∶1、提取温度80 ℃、提取时间80 min,钙提取率为84.21%。 结论 研究结果为大眼金枪鱼骨制备钙制剂提供了理论依据。
Objective In order to fully extract the calcium of fish bones and make high-value utilization of fish bone resources. Method Bigeye tuna (Thunnus obesus) bones are used as raw materials to extract soluble calcium by hydrochloric acid.The calcium content in bigeye tuna bones is measured by high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICPMS).Atomic absorption spectrometry was used to determine the calcium content in the extraction.Based on the optimization of hydrochloric acid concentration,liquid to material ratio,extraction temperature and extraction time in one-factor experiments,the three factors of hydrochloric acid concentration,liquid to material ratio and extraction time were further optimized by Box-Behnken design.The model feasibility analysis was carried out,and the actual value of calcium extraction rate was compared with the predicted value obtained by multiple regression fitting. Result The equations fitted by Box-Behnken could be used to simulate actual conditions,and the modified process parameters were hydrochloric acid concentration 1.00 mol/L,liquid to material ratio 14∶1,extraction temperature 80 °C and extraction time 80 min,calcium extraction rate was 84.21%. Conclusion This study provided a theoretical basis for the calcium preparation from bones of bigeye tuna.
