饲粮中添加不同水平花椒籽对育肥猪脂肪酸组成和FABP1基因组织表达的影响

Effects of different supplemental levels of Zanthoxylum bungeanum seeds on fatty acid composition and tissue expression of the FABP1 gene in fattening pigs
目的 探究饲粮中添加不同水平花椒籽对育肥猪脂肪酸组成和肝脏型脂肪酸结合蛋白基因(FABP1)组织表达水平的影响。 方法 试验选择288头3月龄体质量相当且健康的杜×长×大三元育肥猪,随机分为4组,分别用花椒籽代替基础饲粮中0%(对照组)、2.5%(Ⅰ组)、5%(Ⅱ组)和7.5%(Ⅲ组)玉米进行饲喂,试验期100 d,试验结束后采集育肥猪肝脏、背肌和肾周组织测定脂肪酸含量,利用荧光定量qPCR检测FABP1基因在育肥猪各组织中的表达特征。 结果 添加花椒籽显著改善育肥猪背肌和肝脏中SFAs、MUFAs和PUFAs水平,试验I组背肌和肝脏SFAs显著升高(P<0.05),试验Ⅱ组背肌、肝脏和肾周PUFAs显著升高(P<0.05);猪FABP1基因CDS全长383 bp,共编码127个氨基酸,与参考序列相比,其编码区第53位T突变为A;FABP1基因在花椒籽饲喂育肥猪的肝、脾、十二指肠、空肠、回肠、结肠、直肠、盲肠和背肌组织均显著上调表达(P<0.05),在肾脏中显著下调表达(P<0.05),试验II组脾脏、空肠、直肠和背肌中FABP1基因的表达量显著高于其他组(P<0.05)。 结论 饲粮中添加花椒籽显著改善育肥猪脂肪酸组成,提高各组织FABP1基因的表达量,这有利于育肥猪的脂肪沉积和肉质改善。
Objective This study aimed to investigate the fatty acid composition and tissue expression levels of the liver type fatty acid binding protein gene(FABP1) in fattening pigs fed different levels of Zanthoxylum bungeanum seeds. Method A total of 288 healthy three⁃month⁃old Duroc×Landrace×Yorkshire fattening pigs with the same body weight were randomly selected and divided into 4 groups fed the basal diet with Zanthoxylumbungeanum seeds replacing 0%(control group),2.5%(group Ⅰ),5%(group Ⅱ) and 7.5%(group Ⅲ) corn,respectively.The experiment lasted 100 days.At the end of the experiment,the liver,back muscle and perirenal tissues of the fattening pigs were collected to test the fatty acid content,and the expression characteristics of the FABP1 gene of the fattening pigs were also detected by RT-qPCR. Result The levels of SFAs,MUFAs and PUFAs in back muscle and liver of fattening pigs were significantly improved by dietary supplementation with Zanthoxylum bungeanum seeds,and SFAs of back muscle and liver were significantly increased in group I than that in others (P<0.05),PUFAs of back muscle,liver and perirenal were significantly increased in group II than that in others (P<0.05).The CDS of the FABP1 gene was 383 bp in length and encoded 127 amino acids,compared with the reference sequence,the T in the 53rd position of the coding region was mutated to A,resulting in the missense mutation.The expression levels of FABP1 gene were significantly upregulated in liver,spleen,duodenum,jejunum,ileum,colon,rectum,cecum and back muscle of fattening pigs after feeding Zanthoxylum bungeanum seeds (P<0.05),while significantly downregulated in kidney (P<0.05).The expression levels of FABP1 gene were significantly higher in spleen,jejunum,rectum and back muscle than that in other tissues (P<0.05). Conclusion The addition of Zanthoxylumbungeanum seeds could significantly improve fatty acid composition and increase tissue expression of the FABP1 gene in fattening pigs,which is beneficial for fat deposition and meat quality improvement in finishing pigs.
