甘农4号小黑麦苗期抗寒性评价
Evaluation on cold resistance of Triticosecale variety Gannong No.4 at seedling stage
目的 研究甘农4号小黑麦品种的抗寒性。 方法 以甘农4号为试验材料,石大1号(CK1)和中饲1048(CK2)为对照,利用超低温冰箱人工模拟低温,研究了不同低温胁迫下(0 ℃、-10 ℃、-20 ℃和-30 ℃)3个小黑麦品种的生理生化指标(叶片相对含水量(RWC)、脯氨酸(Pro)、可溶性糖(SS)、可溶性蛋白(SP)、叶绿素(Chl)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性)。 结果 从单因素试验看,甘农4号小黑麦在不同低温胁迫下的平均SS含量显著高于CK2,CAT活性极显著高于CK1和CK2,MDA含量和SOD活性极显著低于CK1和CK2;随着低温胁迫加剧,3个小黑麦品种的平均RWC和Chl含量呈下降趋势,平均MDA含量、SOD,POD和CAT活性逐渐上升,平均Pro、SS和SP含量呈先上升后下降趋势。小黑麦品种×低温胁迫天数的交互表明,0 ℃时,甘农4号小黑麦的RWC和MDA含量,以及SOD和CAT活性显著低于两个对照,Pro含量略高于2个对照,Chl含量略低于2个对照;-10 ℃时,其Pro含量和CAT活性显著高于2个对照;-20 ℃时,其Pro和SS含量及CAT活性显著高于2个对照,MDA含量低于2个对照;-30 ℃时,其Pro含量及POD和CAT活性显著高于两个对照。 结论 甘农4号小黑麦苗期的抗寒性为中等,与CK1相近,强于CK2。
Objective The aim of this study was to evaluate the cold resistance of triticale variety Gannong No.4. Method In this study,the variety Gannong No.4 was employed as the target materials,with the Shida No.1 (CK1) and Zhongsi 1048(CK2) as the control.The physiological and biochemical characteristics of the 3 triticale varieties,including leaf relative water content (RWC),proline (Pro),soluble sugar (SS),soluble protein (SP),chlorophyll (Chl),malondialdehyde (MDA) content and superoxide dismutase (SOD),Peroxidase (POD) and catalase (CAT) activity,were determined under different low-temperature stress (i.e.,0 ℃,-10 ℃,-20 ℃ and -30 ℃). Result Our results showed that when only single factor is considered,the averaged SS content of Gannong No.4 under different low temperature stresses was significantly higher than that of CK2 at the 0.05 level,CAT activity was significantly higher than that of CK1 and CK2 at the 0.01 level,MDA content and SOD activity were significantly lower than that of the CK1 and CK2 at the 0.01 level; With the intensity of the low temperature stress,the averaged RWC and Chl contents of 3 triticale varieties downward the averaged MDA content,activities of SOD,POD and CAT gradually increased,and the averaged contents of Pro,SS and SP firstly increased and then decreased.The interaction of the low temperature stress days and triticale varieties showed that,at 0 ℃,contents of RWC and MDA,and the activities of SOD and CAT of Gannong No.4 triticale were significantly lower than the 2 controls,Pro content was slightly higher than the 2 controls,and the Chl content was slightly lower than the 2 controls;at -10 ℃,its Pro content and CAT activity were significantly higher than 2 controls; at -20 ℃,its Pro and SS content and CAT activity were significantly higher than 2 controls,MDA content was lower than 2 controls; at -30 ℃,its Pro content and POD and CAT activity Significantly higher than 2 controls. Conclusion Comprehensive evaluation showed that the cold resistance of Gannong No.4 at the seedling stage was moderate,which was similar to the CK1 but stronger than the CK2.
