富含羟基酪醇橄榄油对衰老小鼠的影响
Effect of olive oil containing hydroxyl tyrosol on aging mice
目的 为研究富含羟基酪醇橄榄油(Olive oil rich in hydroxyl tyrosol,OHT)对衰老小鼠的影响。 方法 利用小鼠颈部皮下注射500 mg/kg的D-半乳糖(D-galactose)溶液构建亚急性衰老模型,建模成功后,将小鼠随机分成7组,分别为空白对照组(CK0)、模型组(Model group)、维生素E阳性对照组(VE positive control group)、普通橄榄油组(Cormal olive oil group)和OHT高、中、低剂量(200、100、50 mg/kg)处理组(OHT high,medium,low-dose group),连续灌胃28 d,检测小鼠体质量变化及脏器指数、血清中谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)活性及丙二醛(malondialdehyde,MDA)含量的变化,观察肝组织形态学变化。 结果 OHT使衰老小鼠的体质量和脏器指数显著升高(P<0.05,P<0.01),肝脏病理变化得到改善,血清CAT、GSH-Px和SOD活性显著升高(P<0.05,P<0.01),MDA含量显著降低(P<0.05,P<0.01),以上指标变化在OHT高剂量组尤为明显,且OHT干预效果强于普通橄榄油。 结论 OHT能提高机体抗氧化能力,进而发挥抗衰老的作用。
Objective To study the effects of olive oil containing hydroxyl tyrosol (OHT) on aging mice. Method An aging animal model was established by subcutaneous injection of 500 mg/kg D-galactose in the neck,and after successful modeling,the mice were randomly divided into 7 groups: blank control group,model group,VE positive control group,common olive oil group,OHT high,medium and low dose treated groups (200 mg/kg,100 mg/kg,50 mg/kg),the OHT was administered to the mice by intragastric administration for 28 consecutive days.The weight and viscera indexes of each group were calculated,and the activities of glutathione peroxidase (GSH-Px),superoxide dismutase (SOD),catalase (CAT) and malondialdehyde (MDA) in the serum were determined,and the morphology changes of the liver tissue were observed. Result OHT could significantly improve the body weight and organ index in aging mice(P<0.05,P<0.01),ameliorate the pathological changes of the liver,significantly increase the activity of serum CAT,GSH-Px and SOD (P<0.05,P<0.01),and significantly reduced MDA content (P<0.05,P<0.01).The changes in the above indexes were particularly evident in the high-dose OHT group,and the intervention effect of OHT was stronger than that of normal olive oil. Conclusion OHT could improve the body’s antioxidant capacity to play an anti-aging role.
