壳寡糖对冬小麦幼苗抗冷性的影响

壳寡糖对冬小麦幼苗抗冷性的影响
Effect of chito-oligosaccharide on cold resistance of winter wheat
目的 探究壳寡糖对冬小麦幼苗低温胁迫的缓解机制及最适浓度。 方法 以盆栽土培法培养冬小麦(东农冬麦1号和济麦22),利用光照培养箱设置低温环境对两种小麦进行低温胁迫,以去离子水为对照,研究了3个浓度(50、100、150 mg/mL)壳寡糖对小麦生长状况、MDA含量、抗氧化酶活性以及渗透调节物质的影响。 结果 低温胁迫下喷施壳寡糖溶液对东农冬麦1号和济麦22幼苗生长均有促进作用,以100 mg/L壳寡糖效果最明显;与对照相比,喷施50、100 mg/L壳寡糖后东农冬麦1号MDA含量显著降低,分别降低了28.69%和39.06%,济麦22无显著变化;不同浓度的壳寡糖使两种冬小麦的抗氧化酶活性和渗透调节物质含量提高,以喷施100 ml/L壳寡糖效果最好,与对照相比东农冬麦1号和济麦22的SOD活性分别增加了114.56%、102.37%,POD活性增加了39.89%和37.90%,CAT活性增加了84.93%和68.74%,可溶性糖含量增加了142.10%和134.21%,可溶性蛋白增加了11.16%和4.95%,脯氨酸含量增加了457.53%和503.20%。 结论 壳寡糖提高冬小麦抗冷性的最佳喷施浓度为100 mg/L。低温胁迫下喷施适宜浓度的壳寡糖能促进冬小麦幼苗生长,冬小麦通过降低膜脂过氧化程度,提高抗氧化酶活性以及渗透调节物质来增强抗寒能力,且对抗寒性强的东农冬麦1号效果更好。
Objective To explore the mitigation mechanism and optimal concentration of Chito-oligosaccharide on winter wheat seedlings under low temperature stress. Method The cultivars of Dongnong dongmai 1 and Jimai 22 were subjected to low temperature stress in polt soil by light incubator.To explore the effects of different concentrations of chito-oligosaccharide solution(50 mg/L、100 mg/L、150 mg/mL)on the growth,MDA contents,antioxidant enzyme activity and osmosis-regulating substances contents of winter wheat. Result The results showed that spraying 3 concentrations of chito-oligosaccharide could promote growth of Dongnong dongmai 1 and Jimai 22 seedlings under low temperature stress and the growth increased significantly of 100 mg/L chito-oligosaccharide.Foliar-spraying 100 mg/L chito-oligosaccharide significantly decreased the MDA content of Dongnong dongmai under low temperature stress and the MDA content of Dongnong dongmai decreased by 39.06%,the MDA content of Jimai 22 decreased without significant change.Spraying 3 concentrations of chito-oligosaccharide could promote antioxidant enzyme activity and osmosis-regulating substances contents of two cultivars winter wheat under low temperature stress.100 mg/L chito-oligosaccharide has the best effect,the SOD activity of Dongnong dongmai 1 and Jimai 22 increased by 114.56%、102.37%,the POD activity increased by 39.89%、37.90%,the CAT activity increased by 84.93%、68.74%,the soluble sugar increased by142.10%、134.21%,the soluble protein increased by11.16%、4.95% and proline contents increased by 457.53%、 503.20%. Conclusion The best spraying concentration of chito-oligosaccharide is 100 mg/L.The appropriate spraying concentration of chito-oligosaccharide solution could enhance Dongnong dongmai 1 and Jimai 22 resistance to low temperature stress by promoting growth,decreasing membrane lipid peroxidation,iproving the antioxidant enzyme activity and osmosis-regulating substances contents.And chito-oligosaccharide had better effect on Dongnong dongmai 1 with strong cold resistance.
