藏药蓝玉簪龙胆的种子萌发和愈伤繁殖特性

藏药蓝玉簪龙胆的种子萌发和愈伤繁殖特性
Characteristics of seed germination and callus propagation of Tibetan medicine Gentiana veitchiorum
目的 为保护蓝玉簪龙胆的种质资源,探索其人工繁育方法。 方法 试验采用浸种和组织培养技术,通过不同培养基和不同激素的筛选,讨论蓝玉簪龙胆种子萌发以及愈伤组织分化的最佳策略。 结果 蓝玉簪龙胆种子在200 mg/L GA3处理4 h发芽率最高,为44.84%;在不同基本培养基中培养,B5培养基萌发效果最佳,发芽率达53.21%。以B5培养基为基础,向培养基中添加不同浓度的GA3、NAA和6-BA,得到蓝玉簪龙胆种子萌发的最优培养基为:B5+0.5 mg/L GA3+1 mg/L6-BA,发芽率达66.01%;诱导愈伤组织生长的最适宜培养基为:B5+0.5 mg/L GA3+0.5 mg/LNAA+0.5 mg/L6-BA,诱导率为34.28%。以愈伤组织生长最适培养基为基础,添加不同浓度6-BA,进一步优化培养基配方,得到最适合蓝玉簪龙胆愈伤组织继代培养的方案:B5+0.5 mg/LGA3+0.5 mg/LNAA+1 mg/L6-BA。 结论 愈伤组织继代培养方案可以将蓝玉簪龙胆从种子分化成愈伤组织,再分化成幼苗。
Objective The study aimed to explore artificial breeding approaches of the Gentiana veitchiorum in order to protect its germplasm resources. Method The best conditions for seed germination and callus differentiation of G.veitchiorum was determined by using seed soaking and tissue culture techniques and screening of different media and different hormones in the experiment. Result The germination rate of G.veitchiorum seeds was the highest when treated with 200 mg/L GA3 for 4 h, which was 44.84%.Cultured in different basic media, the seeds had the highest germination rate on B5 medium (i.e., 53.21%).With adding different concentrations of GA3, NAA and 6-BA to basic B5 medium, it was showed that the optimal medium for seed germination of Gentiana veitchii was B5+0.5 mg/L GA3+1 mg/L 6-BA,on which the seed germination rate was 66.01%.The optimal medium for callus induction was B5+0.5 mg/L GA3+0.5 mg/L NAA+0.5 mg/L 6-BA,on which the induction rate was 34.28%.Based on the optimal medium for callus growth, the medium formula was further optimized by adding different concentrations of 6-BA,and the most suitable scheme for callus subculture was obtained as follows: B5+0.5 mg/L GA3+0.5 mg/L NAA+1 mg/L 6-BA. Conclusion The G.veitchiorum seeds can be differentiated into callus,and then into seedlings by using the callus subculture scheme.
