季节对海狸色公獭兔血液性激素水平和精液品质的影响
Seasonal influences of sexual hormones and sperm qualities to male beaver Rex rabbit
目的 研究海狸色公獭兔血液中性激素和精液品质随季节变化的规律。 方法 选择36只3月龄公兔做为试验样本,于2020年12月~2021年11月分别采集12月(3月龄)、2月(5月龄)、4月(7月龄)、6月(9月龄)、8月(11月龄 )的颈静脉血样本5.0mL,经ELISA方法测定血清中促卵泡素(FSH)、促黄体素(LH)和睾酮(T)的变化情况。公兔精液从2月(5月龄)开始同步采集,检测精子活力、精子畸形率、精子密度。 结果 性激素水平分析表明:卵泡刺激素(FSH)在2月份达到峰值,极显著高于其它各月(P<0.01);促卵泡素(LH)在4月份达到峰值,极显著高于其它各月(P<0.01);睾酮(T)在2月和6月龄达到峰值,极显著高于其它各月(P<0.01)。精子活力2月份最高(0.59±0.034),4月(0.56±0.013)、6月(0.49±0.011)、8月(0.37±0.008)逐渐下降,各月之间差异极显著(P<0.01);精子畸形率2月(0.065 7±0.008 2)、4月(0.059 0±0.004 6)、6月(0.061 7±0.005 0)极显著低于8月(0.114 2±0.004 9)(P<0.01);精子密度2月(1.88±0.11亿个/毫升)极显著高于4月(1.33±0.12亿个/毫升,P<0.01)和6月(1.54±0.12亿个/毫升,P<0.01),显著高于8月(1.64±0.14亿个/毫升,P<0.05)。相关性分析显示:2月FSH浓度与2月(γ=0.477)、4月(γ=0.352)、6月(γ=-0.261)T值浓度呈显著或极显著的相关性(P<0.05,P<0.01);4月LH浓度与4月T浓度(γ=0.513)的相关性最高;4月LH的浓度与4月T的浓度呈现显著负相关性(γ=-0.295)(P<0.05);12月龄的FSH水平与4月龄精子密度呈极显著相关(P<0.01);12月龄LH水平与6月和8月龄精子密度呈极显著(P<0.01)和显著(P<0.05)相关;6月龄T水平与4月龄精子密度呈显著相关(P<0.05)。 结论 公兔一年四季均可配种使用,最佳配种季节为春季和秋冬季,且FSH、LH和T的分泌具有协同作用,并影响了精子密度。
Objectives In order to study seasonal variation of sexual hormones of male Rex rabbit. Method 36 two- month-old male Rex rabbits were selected and 5.0mL of their jugular blood from December 2020 to November 2021 were collected,and the changing rules of follicle stimulating hormone (FSH),luteinizing hormone (LH),testone (T) were measured using ELISA method. Result The results showed that,FSH level reached the peak at February,significantly higher than other months (P<0.01); LH level reached the peak at April,significantly higher than other months (P<0.01); T level showed two peaks at February and June respectively,both were significantly higher than other months (P<0.01).The sperm motility was highest at February (0.59±0.034),and gradually reduced at April (0.56±0.013),June(0.49±0.011) and August(0.37±0.008),with the significant differences among seasons (P<0.01); sperm deformity rate at February (0.0657±0.0082),April(0.059±0.0046),June(0.0617±0.0050) were significantly lower than August(0.1142±0.0049) (P<0.01); sperm density at February((1.88±0.11)×108/mL) was strongly significantly higher than April((1.33±0.12)×108/mL),June((1.54±0.12)×108/mL)(P<0.01),significantly higher than August((1.64±0.14)×108/mL) (P<0.05).The correlation analysis showed that FSH level at February was significantly correlated with T level at February(γ=0.477),April(γ=0.352)and June(γ =-0.261)(P<0.05,P<0.01); LH level at April was significantly correlated with T level at April(γ=0.513)(P<0.05); and LH level at April was negatively correlated with T level at April(γ=-0.295)(P<0.05).The FSH level at December was significantly correlated with sperm density at April (P<0.01); and the LH level at December were strongly significantly correlated with sperm density at June (P<0.01),significantly correlated with sperm density at August (P<0.05); the T level at June were significantly correlated with sperm density at April (P<0.05).This study showed that the best breeding season for male rabbit was spring,the worst mating season was summer and beginning of autumn,and the secretion of FSH,LH and T had synergy,which were influenced sperm density. Conclusions This study proved that male rabbit can breed all seasons,and the optimal mating season is Autumn and Spring,and the secretion of FSH,LH and T were in coordination,and influenced the density of sperm.
