丹元糖痹乳膏对小鼠的镇痛和活血化瘀作用的效果评价
Evaluation of analgesic and promoting blood circulation effects of a compound preparation of DYTB Cream in mice
目的 探讨丹元糖痹乳膏(DYTB)对小鼠镇痛和活血化瘀作用,并应用网络药理学探讨其作用机理。 方法 通过小鼠热刺痛、甲醛致痛模型评价DYTB对小鼠镇痛作用;通过耳廓微循环实验,判断DYTB活血作用的功效。采用TCMSP数据库检索DYTB活性成分潜在靶点并通过GeneCards数据库获取疾病相关靶基因。最后应用分子对接技术,检验DYTB主要组分与靶基因之间的结合活性。 结果 小鼠药效试验提示,与模型组比较,DYTB工艺1、2组对神经和/或炎性疼痛均具有显著镇痛效果和促进微循环作用(P<0.01或0.05)。通过网络分析获取DYTB治疗疼痛和血栓的主要活性成分为槲皮素、木犀草素、大黄素和β-谷甾醇等,潜在关键靶点包括IL6、TNF、IL1B和AKT1等。分子对接结果提示,主要成分与关键靶点可有效结合。 结论 DYTB乳膏具有镇痛和活血化瘀作用,为后续DYTB乳膏的研究提供试验研究依据。
Objective To study the effects of Danyuan Tangbi (DYTB) Cream on analgesia and promoting blood circulation in mice,and to explore its mechanism by network pharmacology. Method The effects of DYTB were evaluated on analgesia using heat prickle and formaldehyde-induced pain models in mice.The efficacy of DYTB in promoting blood circulation was assessed by studying auricular microcirculation.Potential targets of DYTB's active components were searched using the TCMSP database,and pain and thrombosis targets were obtained using the GeneCards database.Finally,molecular docking technology was used to test the binding activity between DYTB components and target genes. Result The pharmacodynamic tests showed that the process of DYTB process 1 and 2 group had significant analgesic effect on nerve and/or inflammatory pain and microcirculatory promotion (P<0.05 or 0.01) in mice.Through network analysis,the main active components of DYTB were quercetin,daidzein,emodin and bata-sitosterol,etc.,and the potential key targets included IL6,TNF,IL1B and AKT1,etc.in the treatment of pain and thrombosis.The molecular docking results indicated that the key components could effectively bind to the key targets. Conclusion DYTB Cream has analgesic and circulatory promoting effects which provide an important test base for further research on DYTB Cream.
