冷季不同饲养方式下牦牛皮下脂肪转录组分析
Transcriptome analysis of subcutaneous fat in yaks under different feeding regimes during the cold season
目的 旨在探究牦牛冬季自然放牧和舍饲条件下皮下脂肪转录表达差异。 方法 以9头18月龄体质量相近、背部皮下脂肪差异不显著的牦牛为试验对象,随机分为3组,于试验初期(10月)选取对照组(G18)牦牛进行屠宰,试验组1(G24)继续放牧6个月,试验组2(F24)舍饲6个月分别屠宰,采集3组背部皮下脂肪进行转录组测序、差异表达基因筛选以及富集分析。 结果 本研究获得了9个cDNA文库,差异表达分析结果显示,在G18vsF24中检测到上调基因160个,下调基因249个,其中有309个差异基因富集到262个通路中,主要有PPAR信号通路、甘油酯代谢、AMPK信号通路等;在G18vsG24中检测到上调基因615个,下调基因361个,其中有735个差异基因富集到317个通路中,细胞因子-细胞因子受体相互作用、不饱和脂肪酸生物合成等;在F24vsG24中检测到上调基因161个,下调基因91个,其中有188个差异基因富集到249个通路中,趋化因子信号通路、细胞因子-细胞因子受体相互作用和甘氨酸、丝氨酸及苏氨酸代谢等。 结论 本研究结果揭示了不同饲养方式基因差异表达、富集通路信息的不同,为进一步深入探究牦牛皮下脂肪的分子调控以及对牦牛脂肪沉积的研究提供了参考。
Objective The aim of this experiment was to investigate the differences in transcript expression in subcutaneous fat of yaks under natural grazing and winter housing conditions. Method Nine 18-month-old yaks with similar body weight and non-significant differences in back subcutaneous fat were randomly divided into three groups,and the control group (G18) was slaughtered at the beginning of the experiment (October),the test group 1 (G24) continued to graze for 6 months,and the test group 2 (F24) was housed for 6 months,three groups of back subcutaneous fat were collected for transcriptome sequencing,differentially expressed gene screening and enrichment analysis. Result Nine cDNA libraries were obtained,and differential expression analysis showed that 160 up-regulated genes and 249 down-regulated genes were detected in G18vsF24,of which 309 differential genes were enriched in 262 pathways,mainly PPAR signaling pathway,glycerol ester metabolism,AMPK signaling pathway,etc.;615 up-regulated genes and 361 down-regulated genes were detected in G18vsG24 361 down-regulated genes were detected in G18 vs.G24,of which 735 differential genes were enriched into 317 pathways,cytokine-cytokine receptor interaction,unsaturated fatty acid biosynthesis,etc.;161 up-regulated genes and 91 down-regulated genes were detected in F24 vs.G24,of which 188 differential genes were enriched in 249 pathways,chemokine signaling pathway,cytokine-cytokine receptor interaction,and glycine,serine and threonine metabolism,among others. Conclusion The results reveal the differences in gene differential expression and enrichment pathway information in different feeding regimes,and the results provide a reference for further in-depth investigation into the molecular regulation of subcutaneous fat in yaks and the study of fat deposition in yaks.
