女黄扶正提取液体外抗羊痘病毒活性及对小鼠免疫功能的影响

女黄扶正提取液体外抗羊痘病毒活性及对小鼠免疫功能的影响
In vitro anti-GTP virus activity of the Nyuhuang Fuzheng extraction solution and its effect on the immune function of mice
目的 研究女黄扶正提取液体外对羊痘病毒( goat pox virus,GTPV)的抑制作用及其机制,以及调节小鼠免疫功能的作用。 方法 测定女黄扶正提取液对羊肾细胞的毒性、确定最佳的用药浓度,分别测定病毒感染12、24、36、48 h后病毒拷贝数和TCID50。通过qPCR分析药物处理组、对照组、空白组的细胞炎性因子IL-1β、TNF-α、IFN-β表达量。检测给药后免疫抑制小鼠血清中免疫指标及脾T淋巴细胞转化能力。 结果 女黄扶正提取液对羊肾细胞存在一定细胞毒性,在添加量为1 mg/mL时对细胞毒性最小,感染羊痘病毒12、24、36、48 h后,病毒拷贝数低于对照组,在48 h呈现显著性差异(P<0.05);病毒TCID50低于对照组,在24、48 h呈现显著性差异(P<0.05、P<0.01)。感染病毒后的药物处理组与对照组的细胞炎性因子IL-1β、TNF-α、IFN-β的表达量无显著性差异。给药后,女黄扶正提取液中剂量组小鼠脾T淋巴细胞转化能力明显升高,与模型组相比,差异极显著(P<0.01);免疫抑制小鼠血清中IFN-γ、IL-2升高,与模型组相比,差异极显著(P<0.01)。 结论 女黄扶正提取液可以体外抑制羊肾细胞中GTPV的增殖,提高小鼠IFN-γ、IL-2水平,调节机体免疫功能,为提高机体免疫及未来抗病毒药物的研发提供参考。
Objective The study aimed to explore the inhibitory effect of Nyuhuang Fuzheng extract on goat pox virus (GTPV) in vitro and the mechanism,and its immunomodulatory effect on mice. Method To determine the toxicity of the extract to sheep kidney cells and the optimal drug concentration,and the virus copy number and TCID50 were determined 12,24,36,48 h after virus infection.The expression levels of inflammatory factors IL-1β,TNF-α and IFN-β were analyzed in the drug treatment,the control and the blank group by qPCR.The serum immune indexes and splenic T lymphocyte transformation ability of immunosuppressed mice were detected after administration. Result Nyuhuang Fuzheng extract had a certain cytotoxicity to sheep kidney cells,and the cytotoxicity was the least when the added amount was 1 mg/mL.At 12,24,36,48 h after infection,the virus copy number was lower than that of the control group,and the difference was significant at 48 h (P<0.05).TCID50 of the virus was also lower than the control group,and the difference showed the significance at 24 h and 48 h (P<0.05,P<0.01).There was no significant difference in the expression of IL-1β,TNF-α and IFN-β after infection between the drug treatment and the control group.Compared with the model group,the transformation ability of spleen T lymphocytes in the middle-dose group was significantly increased (P<0.01).The levels of IFN-γ and IL-2 in the serum of immunosuppressed mice were significantly higher than those in the model group (P<0.01). Conclusion The Nyuhuang Fuzheng extract could inhibit the proliferation of GTPV in sheep kidney cells in vitro,increase the levels of IFN-γ and IL-2 in mice,and regulate the immune function of the organism,which provided a reference for improving immunity and the development of future antiviral drugs.
