牡丹果荚的质量标准研究
Study on the quality standard of the pods of Paeonia ostii T. Hong & J. X. Zhang
目的 建立牡丹果荚的质量标准。 方法 用高效液相色谱法(high performance liquid chromatography,HPLC)测定牡丹果荚中没食子酸、鞣花酸的含量;用薄层色谱法(thin layer chromatography,TLC)鉴别牡丹果荚中没食子酸;按照2020年版《中华人民共和国药典》中的方法测定水分、总灰分、酸不溶性灰分、水溶性浸出物的含量;用生物显微镜观察法对果荚粉末进行鉴别。 结果 建立了专属性强、重复性好的药材中没食子酸的TLC鉴别方法;建立了药材中没食子酸及鞣花酸含量测定方法:没食子酸在1.30~166.70 μg·mL-1范围内线性关系良好(r=0.999 5),平均加样回收率为96.66%,RSD值为1.95%;鞣花酸在1.00~128.26 μg·mL-1范围内线性关系良好(r=0.999 3),平均加样回收率为98.22%,RSD值为1.15%;12批药材中没食子酸、鞣花酸、水分、总灰分、酸不溶性灰分、水溶性浸出物的质量分数分别为7.73~10.00 mg·g-1、11.31~12.50 mg·g-1、9.98%~11.07%、10.35%~12.35%、0.32%~0.41%、32.27%~33.55%;果荚粉末观测到簇晶、导管和非腺毛。 结论 所建立的牡丹果荚质量标准可用于该药材的质量控制。
Objective To establish the quality standard of the pods of Paeonia ostii T. Hong & J. X. Zhang. Methods High performance liquid chromatography (HPLC) was used to determine the contents of gallic acid and ellagic acid in the pods of Paeonia ostii T. Hong&J. X. Zhang. Thin layer chromatography (TLC) was used to identify gallic acid. The contents of water, total ash, acid insoluble ash and water-soluble extract were determined according to Chinese Pharmacopoeia (2020 Edition). The powders of the pods were identified by biological microscope. Results A TLC identification method of gallic acid with strong specificity and good repeatability was established. Methods for the determination of gallic acid and ellagic acid was also established. Gallic acid has a good relationship in the linear range of 1.30—166.70 μg·mL-1 with regression coefficient r=0.999 5, and its average recovery was 96.66% with RSD 1.95%. The ellagic acid has a good relationship in the range of 1.00—128.26 μg·mL-1 with regression coefficient r=0.999 3, and its average recovery was 98.22% with RSD 1.15%. The mass fraction of gallic acid, ellagic acid, water, total ash, acid insoluble ash and alcohol soluble extract in 14 batches of the pods were 7.73—10.00 mg·g-1, 11.31—12.50 mg·g-1, 9.98%—11.07%, 10.35%—12.35%, 0.32%—0.41%, and 32.27%—33.55%, respectively. Tufts, catheters and non-glandular hairs can be observed in the powder. Conclusion The established HPLC method, TLC method and microscopic method can be used for the qua⁃lity control of the pods of Paeonia ostii T. Hong & J. X. Zhang.
