正天丸中4种微量元素的含量测定及健康风险评估
Determination of 4 trace elements in Zhengtian Pills and health risk assessment
目的 建立测定正天丸中铅(Pb)、砷(As)、镉(Cd)、铬(Cr)4种微量元素含量的方法,并对其进行健康风险评估。 方法 采用微波消解法对正天丸样品进行处理,采用等离子体发射光谱法(inductively coupled plasma atomic emission spectrometry,ICP-AES)检测正天丸中4种微量元素的含量,利用聚类分析和相关性分析对数据进行处理;采用靶向危害系数法(target hazard quotient,THQ)和“中药有害物质限量制定指导原则”中的重金属及有害元素最大限量法对不同批次正天丸样品中4种微量元素的污染程度进行风险评估。 结果 在20.0~200.0 ng·mL-1质量浓度范围内,4种元素的峰面积与质量浓度呈良好线性关系(r≥0.999 7),检出限为0.52~1.14 ng·mL-1;加样回收率为98.62%~101.83%,精密度实验、重复性实验、加标回收率实验的相对标准偏差(relative standard deviation,RSD)均小于3%。正天丸中4种微量元素的含量有差异但均符合《中华人民共和国药典》(2020版)中的相关要求,对人体健康不会产生明显影响。 结论 该方法准确度高、精密度好、分析速度快,可同时测定多种元素,适用于正天丸中4种微量元素的测定,检测的正天丸质量安全性较好,为正天丸的健康风险评估提供科学依据。
Objective To establish a method for the determination of the 4 trace elements (Pb, As, Cr, Cd) in Zhengtian Pills, and to assess its health risk. Methods The samples were treated by nitric acid microwave digestion system. Microwave digestion-inductively coupled plasma atomic emission spectrometry (ICP-AES) method was established to determine the content of the 4 trace elements. The results were analyzed by using cluster analysis and correlation analysis. Risk assessment of the 4 trace elements pollution levels in different batches of Zhengtian Pills was performed by using targeted hazard quotients (THQ) and the maximum limit method of heavy metals and toxic elements in “Guideline for Formulation of the Limit of toxic Residue of Traditional Chinese Medicine”. Results The peak areas of the 4 elements showed a good linear relationship with the mass concentration within the 20.0-200.0 ng·mL-1 concentration range (r≥0.999 7). The limits of detection were in the range of 0.52-1.14 ng·mL-1 and the recoveries were in the range of 98.62%-101.83%. The RSD of precision test, repeatability test, and spiked recovery rate test were all less than 3%. There were differences in the content of the 4 trace elements in Zhengtian Pills, but they complied with the relevant requirements of the Chinese Pharmacopoeia(2020 edition). The 4 trace elements in Zhengtian Pills did not show significant impact on human health. Conclusion The established method has the chracteristics of high accuracy, good precision, and fast analysis speed, which make the simultaneous determination of multiple elements be possible. The quality of the tested Zhengtian Pills is generally safe.
