一测多评法同时测定润肠丸(胶囊)中的9种成分
Determination of 9 constituents in Runchang Pills (Capsules) by quantitative analysis of multi-components with single-marker
目的 建立同时测定润肠丸(胶囊)中苦杏仁苷、阿魏酸、芦荟大黄素、大黄酸、羌活醇、大黄素、异欧前胡素、大黄酚、大黄素甲醚含量的一测多评法(quantitative analysis of multi-components with single-marker,QAMS)。 方法 该药物甲醇提取液的分析采用Agilent Venusil C18 Plus色谱柱(4.6 mm × 250 mm,5 μm);流动相为乙腈(A)-体积分数0.1%磷酸(B)-水(C);梯度洗脱;柱温为30 ℃;检测波长为218 nm(苦杏仁苷)、254 nm(阿魏酸、芦荟大黄素、大黄酸、羌活醇、大黄素、异欧前胡素、大黄酚、大黄素甲醚);以芦荟大黄素为内标物,建立与其他8种成分的相对校正因子,检测各成分的含量。 结果 苦杏仁苷、阿魏酸、芦荟大黄素、大黄酸、羌活醇、大黄素、异欧前胡素、大黄酚、大黄素甲醚的质量浓度分别在11.48~229.56、2.79~55.92、2.34~46.96、3.44~68.82、3.01~60.33、2.07~41.49、4.34~86.82、4.69~93.76、3.78~75.72 µg·mL-1范围内有良好的线性关系(r≥0.996 9);加样回收率为97.42%~99.28%,相对标准偏差(relative standard deviation,RSD)为1.09%~2.27%;QAMS结果与外标法实测值无明显差异。 结论 建立的方法准确、可靠,稳定性、耐用性好,可用于润肠丸(胶囊)中苦杏仁苷、阿魏酸、芦荟大黄素、大黄酸、羌活醇、大黄素、异欧前胡素、大黄酚、大黄素甲醚含量的测定。
Objective To establish a quantitative analysis method for simultaneous content determination of amygdalin, ferulic acid, notopterol, isoimperatorin and other 5 anthraquinone derivatives in Runchang Pills (Capsules) by using the analysis of multi-components by single-marker (QAMS). Methods The determination was carried out by using high-performance liquid chromatography (HPLC). An Agilent Venusil C18 Plus column (250 mm×4.6 mm, 5 μm) was used with a mobile phase consisting of acetonitrile, 0.1% phosphoric acid and water in a gradient elution mode. The flow rate was maintained between 0.7~1.0 mL·min-1, the column temperature was set at 30 ℃, and the detection wavelengths were 218 nm and 254 nm respectively. The content was calculated by using QAMS with aloe emodin as an internal standard. Results The mass concentrations of amygdalin, ferulic acid, aloe emodin, rhein, notopterol, emodin, isoimperatorin, chrysophanol, and physcion showed good linear relationships (r≥0.996 9) within the linear ranges of 11.48-229.56, 2.79-55.92, 2.34-46.96, 3.44-68.82, 3.01-60.33, 2.07-41.49, 4.34-86.82, 4.69-93.76 and 3.78-75.72 µg·mL-1 respectively. The recovery rates were ranged from 97.42% to 99.28%, with RSD levels ranging from 1.09% to 2.27%. The results obtained by the 2 quantitative determination methods were consistent. Conclusion This method is accurate, reliable, stable and durable, providing an experimental basis for enhancing the standards of Runchang Pills (Capsules).
