不同氨基酸前体对紫花鄂北贝母培养物及其总生物碱含量的影响
Effects of different amino acid precursors on the growth and total alkaloid content of Fritillaria ebeiensis var. purpurea G. D. Yu et P. Li culture
目的 探讨在紫花鄂北贝母液体培养基中添加不同氨基酸前体对其培养物生长及总生物碱积累的影响,为优化紫花鄂北贝母悬浮细胞培养体系及提高有效成分产量提供实验依据。 方法 采用单因素实验考察色氨酸(tryptophan, Trp)、苯丙氨酸(phenylalanine,Phe)、酪氨酸(tyrosine,Tyr)单一添加时不同浓度对紫花鄂北贝母培养物增殖及总生物碱含量的影响;采用L9(33)三因素三水平正交实验探究3种不同氨基酸组合添加的最优配比;通过紫外-可见分光光度法测定培养物中总生物碱含量。 结果 单一添加条件下,0.6 mmol·L-1 Trp对紫花鄂北贝母培养物生长的促进作用最显著,增殖倍数达2.14;0.4 mmol·L-1 Trp和0.4 mmol·L-1 Phe单独添加时,培养物总生物碱含量均达最大值1.00%。正交实验结果显示,氨基酸组合添加对总生物碱积累的影响主次顺序为Phe>Trp>Tyr,最优组合为0.6 mmol·L-1 Phe+0.4 mmol·L-1 Trp+0.4 mmol·L-1 Tyr,总生物碱含量为0.95%;3种氨基酸对培养物增殖的影响差异无统计学意义(P>0.05)。 结论 添加Trp、Phe、Tyr均可显著提高紫花鄂北贝母培养物的总生物碱含量,其中Trp可促进培养物增殖,而Phe、Tyr单独添加对培养物增殖有一定抑制作用。
Objective To explore the effects of adding different amino acid precursors to the liquid culture medium of Fritillaria ebeiensis var. purpurea G. D. Yu et P. Li on the growth of the culture and the accumulation of total alkaloids. It aims to provide experimental evidence for optimizing the suspension cell culture system of Fritillaria ebeiensis var. purpurea G. D. Yu et P. Li and increasing the yield of active components. Methods A single-factor experiment was conducted to investigate the effects of different concentrations of tryptophan (Trp), phenylalanine (Phe), and tyrosine (Tyr) added singly on the proliferation of the Fritillaria ebeiensis var. purpurea G. D. Yu et P. Li culture and the total alkaloid content; an L9(33) three-factor three-level orthogonal experiment was used to explore the optimal combination ratio of the 3 different amino acids; the total alkaloid content in the culture was determined by ultraviolet-visible spectrophotometry. Results Under the condition of single addition, 0.6 mmol·L-1 Trp showed the most significant promoting effect on the growth of the Fritillaria ebeiensis var. purpurea G. D. Yu et P. Li culture and the proliferation rate reached 2.14; when 0.4 mmol·L-1 Trp and 0.4 mmol·L-1 Phe were added separately, the total alkaloid content of the culture reached the maximum value of 1.00%. The results of the orthogonal experiment showed that the influence of amino acid combinations on the accumulation of total alkaloids was in the order of Phe>Trp>Tyr, and the optimal combination was 0.6 mmol·L-1 Phe + 0.4 mmol·L-1 Trp + 0.4 mmol·L-1 Tyr, the total alkaloid content was 0.95%; there was no statistically significant difference in the effects of the 3 amino acids on the proliferation of the culture (P>0.05). Conclusion Adding Trp, Phe, and Tyr can significantly increase the total alkaloid content of the cultivated Fritillaria ebeiensis var. purpurea G. D. Yu et P. Li. Among them, Trp can promote the proliferation of the culture, while the addition of Phe and Tyr alone has a certain inhibitory effect on the proliferation of the culture.
