MicroRNA-410-3p、KLF6水平与溃疡性结肠炎患者临床特征及严重程度的关系

MicroRNA-410-3p、KLF6水平与溃疡性结肠炎患者临床特征及严重程度的关系
Correlation of microRNA-410-3p and KLF6 levels with clinical characteristics and disease severity in patients with ulcerative colitis
目的 分析microRNA-410-3p(miR-410-3p)、Krüppel样因子6(KLF6)水平与溃疡性结肠炎(UC)患者临床特征及严重程度的关系。 方法 回顾性分析2022年12月—2025年9月新疆维吾尔自治区中医院收治的128例UC患者,并根据疾病严重程度分为缓解期、轻度活动期、中度活动期和重度活动期。另取同期该院健康体检者90例为对照组。通过逆转录聚合酶链反应检测miR-410-3p相对表达量,酶联免疫吸附试验检测KLF6水平。比较UC组与对照组miR-410-3p相对表达量、KLF6水平,以及两者在不同临床特征、不同严重程度UC患者间的差异。 结果 UC组miR-410-3p相对表达量低于对照组,KLF6水平高于对照组(P <0.05)。随着病变范围扩大,患者miR-410-3p相对表达量逐渐降低,KLF6水平逐渐升高(P <0.05)。慢性持续型患者miR-410-3p相对表达量低于慢性复发型及初发型患者(P <0.05),初发型、慢性复发型、慢性持续型患者KLF6水平逐渐升高(P <0.05)。随着严重程度增加,患者miR-410-3p相对表达量逐渐降低(P <0.05)。 结论 血清miR-410-3p相对表达量下降、KLF6水平升高与UC活动度增加、病变范围扩大及慢性进展有关。
Objective To analyze the relationship of the microRNA-410-3p (miR-410-3p) and Krüppel-like factor 6 (KLF6) levels with the clinical characteristics and severity of ulcerative colitis (UC). Methods A retrospective cohort study was conducted on 128 patients with UC admitted to Xinjiang Uygur Autonomous Region Hospital of Traditional Chinese Medicine between December 2022 and September 2025. Patients were categorized into four groups based on disease severity: remission group, mildly active group, moderately active group, and severely active group. Additionally, 90 healthy individuals undergoing routine health check-ups during the same period were selected as the control group. The relative expression of miR-410-3p was detected using reverse transcription polymerase chain reaction (RT-PCR), while KLF6 levels were measured via enzyme-linked immunosorbent assay (ELISA). Comparisons were made between the UC group and the control group, as well as among UC patients with varying clinical characteristics and disease severity, regarding relative expression of miR-410-3p and KLF6 levels. Results The UC group exhibited lower relative expression of miR-410-3p and higher KLF6 levels than the control group (P < 0.05). As the lesion area expanded, the relative expression of miR-410-3p in patients gradually decreased (P < 0.05), while the level of KLF6 gradually increased. The relative expression of miR-410-3p in UC patients with the chronic persistent type was lower than that in patients with the chronic relapsing type and the initial onset type (P < 0.05), whereas the KLF6 level in patients with the initial onset type, chronic relapsing type, and chronic persistent type gradually increased (P < 0.05). With the increase in disease severity, the relative expression of miR-410-3p in patients gradually decreased (P < 0.05). Conclusion Decreased relative expression of serum miR-410-3p and elevated KLF6 levels correlate with increased UC activity, expanded lesion extent, and chronic disease progression.
