摘要
番茄褐色皱纹果病毒(ToBRFV)被列入我国检疫性植物病毒,其严重威胁全球番茄生产。烟草脆裂病毒(TRV)是病毒诱导基因沉默(VIGS)的高效载体,但是否可作为“弱毒疫苗”诱导交叉保护以防控病毒的潜力还有待明确。通过将 ToBRFV 基因组不同长度的片段(正向 100、200、300、500 bp,反向 300 bp)插入 TRV 载体,构建一系列重组 TRV 载体,应用在本氏烟和番茄上,分析重组 TRV 载体的保护效果。结果表明:在本氏烟中,插入正向 300 bp 较反向 300 bp 的重组 TRV 载体有更好的效果,且保护效果随插入正向序列长度增加而增强;在番茄中,插入长度 ≤200 bp 时,保护效果较差。构建的基于 TRV 的 ToBRFV 弱毒疫苗,可为 ToBRFV 防控提供新方案。
