18种石竹属材料叶绿体DNA和线粒体DNA的SSR分析及其与花表型的关联性初探
SSR Analysis of Chloroplast DNA and Mitochondrial DNA of 18 Species of Dianthus and Its Correlation with Floral Phenotypes
植物叶绿体基因组和线粒体基因组均具备母系遗传的特点,可用于明确植物遗传多样性及亲缘关系。中国石竹(Dianthus chinesis)栽培种花瓣花色丰富,本研究采用前期试验中挑选出的100对SSR引物对12种中国石竹栽培种和6种石竹属其他花卉材料的叶绿体DNA和线粒体DNA进行分析。在叶绿体DNA分析中,有30对SSR引物可扩增出多态性条带,在遗传相似系数为0.50时可聚为2个大群:第I大群包括DCPS的1~3、Dca、Dsu、Dba-Sooty、Dba-Red和Dkn,花瓣中均不含有白色部分;第II大群包括SCBS的1~3、Dch-UW、Dch和BCBS的1~5,除Dch外,花瓣均含有白色部分。在线粒体DNA分析中,有28对SSR分析可扩增出多态性条带,在遗传相似系数为0.50时可聚为2个大群:第I大群包括DCPS的1~3、Dca、Dsu、Dch、BCBS的1~5、SCBS的1~3和Dch-UW,均属于大花类;第II大群中Dkn、Dba-Sooty和Dba-Red均为小花类。基于叶绿体DNA和线粒体DNA的SSR分析,中国石竹栽培种间及其与石竹属其他花卉的遗传相似系数分别在0.46~0.73和0.45~0.68。
This study investigated the genetic diversity and relationships of the chloroplast and mitochondrial genomes in plants, focusing on Dianthus chinensis cultivars, which exhibit a wide range of petal colors. A total of 100 SSR primers selected from a previous experiment were used to analyze the chloroplast DNA and mitochondrial DNA of 12 cultivars of Dianthus chinensis, along with 6 other plant species of Dianthus. In the chloroplast DNA analysis, amplified bands were polymorphic by 30 SSR primer pairs. There were two major clusters at a genetic similarity coefficient of 0.50. Cluster I included DCPS from 1 to 3, Dca, Dsu, Dba-Sooty, Dba-Red, and Dkn, had no white sections in their petals. Cluster II comprised SCBS from 1 to 3, Dch-UW, Dch and BCBS from 1 to 5, had white sections in their petals except for Dch. In mitochondrial DNA analysis, amplified bands were polymorphic by 28 SSR pairs. There were two major clusters at a genetic similarity coefficient of 0.50. Cluster I included DCPS from 1 to 3, Dca, Dsu, Dch, BCBS from 1 to 5, SCBS from1 to 3 and Dch-UW, had large sizes of petals. Cluster II consisted of Dkn, Dba-Sooty and Dba-Red, had small sizes of petals. The genetic similarity coefficients among the D. chinensis cultivars ranged from 0.46 to 0.73, while the coefficients between D. chinensis cultivars and other Dianthus species ranged from 0.45 to 0.68, based on SSR analysis of chloroplast and mitochondrial DNA.
