青海省蚕豆赤斑病牡丹葡萄孢病原菌分离鉴定、生物学特性及致病性分化研究

青海省蚕豆赤斑病牡丹葡萄孢病原菌分离鉴定、生物学特性及致病性分化研究
Study on Isolation, Identification, Biological Characteristics and Pathogenicity Differentiation of the Pathogen of Broad Bean Chocolate Spot in Qinghai Province
为明确青海省蚕豆赤斑病病原菌牡丹葡萄孢(Botrytis paeoniae)的生物学特性及其致病能力的相关特征,该研究通过组织分离法从病斑组织中分离并纯化获得目标菌株,综合运用形态学观察、分子生物学鉴定和生物学特性进行分析,并选用青海省12个蚕豆资源进行了致病性评估。形态学特征:菌落边缘规则,菌核呈不规则分布,菌丝呈淡褐色,具二叉分枝结构及隔膜,其分生孢子多呈卵形至梨形,分生孢子梗呈圆形,微着色或近透明。分子鉴定:基于ITS和G3PDH基因序列分析,确认该病原菌为牡丹葡萄孢。生物学特性:牡丹葡萄孢在PDA培养基上生长最快,最适碳源为葡萄糖,氮源以牛肉膏和硝酸钾效果最佳,其最适pH为中性(pH 7),光照条件下以12 h光照/12 h黑暗或24 h连续光照更有利于其生长。致病性分化:不同的蚕豆资源对病原菌的敏感性不同,其中对‘云122’的致病作用最为显著。
To clarify the biological characteristics and pathogenicity of Botrytis paeoniae, the pathogen of broad bean red spot in Qinghai Province, the target strain was isolated and purified from the lesion tissue by using a tissue separation method. The morphological observation, molecular biological identification and biological characteristics were used for analysis, and 12 broad bean resources in Qinghai Province were selected for pathogenicity evaluation. Morphological characteristics revealed that the colony edge was regular, the sclerotium was irregularly distributed, and the mycelium was light brown, with a two-branched structure and a diaphragm. The conidia were mostly oval to rod-shaped, and the conidiophores were round, light brown or nearly transparent. Molecular identification based on ITS and G3PDH gene sequence analysis confirmed the pathogen as Botrytis paeoniae; Biological characteristics indicated that Botrytis paeoniae suffruticosa grew fastest on PDA medium. The optimum carbon source was glucose, and the optimum nitrogen source was beef extract and potassium nitrate. The optimum pH was neutral (pH 7). Under light conditions, 12 h light / 12 h dark or 24 h continuous light was more conducive to growth. Pathogenicity differentiation showed that different faba bean resources had different sensitivities to pathogens, and the pathogenic effect on 'Yun 122 ' was the most significant.
