GH基因在贵州从江香猪不同器官和组织中的表达水平研究
Expression level of the GH gene in different organs/tissues of Congjiang Xiang pigs from Guizhou Province
目的 为解决贵州从江香猪生长性能低、繁殖能力薄弱等问题,合理保护、开发利用优质种质资源。 方法 选取6头1岁左右、体况相近(体重50 kg左右)的贵州从江香猪,分别采集肺、心脏、脾脏、胃、肾脏、肝脏、大肠、脂肪组织、小肠以及背最长肌等组织器官样品并提取总RNA。采用实时荧光定量PCR(qRT-PCR)技术,测定GH基因在的相对表达量。 结果 GH基因在贵州从江香猪检测的所有器官和组织中均有表达,在肝脏的表达水平最高,其次为小肠,且这2个组织中的表达量显著高于其他组织;而背最长肌、肾脏、肺脏、脾脏的表达量显著高于心脏、脂肪、大肠和胃。 结论 GH基因在从江香猪的不同器官和组织中的表达量存在一定差异,未来应进一步研究GH基因在缩短育种时长、优化家畜品种遗传特性方面的作用机制。
Objectives The problems of low growth performance and weak reproductive ability of Congjiang Xiang pigs from Guizhou Province were solved to reasonably protect, develop and utilize germplasm resources with high-quality. Methods 6 Congjiang Xiang pigs from Guizhou Province around 1 year old with similar physical condition( body weight of about 50 kg) were used to extract total RNA from the lungs, heart, spleen, stomach, kidneys, liver, large intestine, adipose tissue, small intestine, and longissimus dorsi muscle. The real-time quantitative PCR technology was used to determine the relative expression levels of the GH gene in these different tissues and organs. Results The GH gene was expressed in all tested organs and tissues of Congjiang Xiang pigs, with the highest level of expression in the liver, followed by in the small intestine. The level of expression in these two tissues was significantly higher than that in other tissues(P<0.05). The level of expression in the longissimus dorsi muscle, kidneys, lungs, and spleen were significantly higher than that in the heart, fat, large intestine, and stomach (P<0.05). Conclusions There were certain differences in the expression level of the GH gene in different organs and tissues of Congjiang Xiang pigs, and the mechanism of the GH gene in shortening the breeding period and optimizing the genetic characteristics should be further studied.
