1株鸡源大肠杆菌的分离鉴定及耐药性分析
Isolation, identification and drug resistance of a strain of chicken-origin E.coli
目的 为确定2024年11月末湖北省随州市某蛋鸡养殖场蛋鸡死亡的病因,并指导临床用药。 方法 采集病死蛋鸡的心脏、肝脏等病变组织样品,通过细菌分离培养、涂片镜检及生化鉴定进行菌落生长特性、形态的观察,通过PCR技术及动物试验检测16S rRNA扩增及攻毒动物生存情况进行测序分析和毒力鉴定,同时对分离菌株进行药敏试验,根据判定标准筛选特效治疗药物,指导临床用药方案。 结果 病死蛋鸡出现明显的包心包肝病理特征,细菌分离培养及鉴定结果均确定该试验分离菌株为致病性大肠杆菌,确诊该病例为大肠杆菌感染,且对氟苯尼考、氯霉素等10种抗生素完全耐药,但对多黏菌素B、氧氟沙星、诺氟沙星、哌拉西林、头孢曲松和头孢噻肟等高度敏感。 结论 分离菌株呈现多重耐药性,药敏试验结果可作为该养殖场的临床治疗用药参考。
Objectives The cause for death of laying hens in a chicken farm in Suizhou City, Hubei Province at the end of November 2024 was determined to guide clinical medication. Methods Samples of diseased tissues including the heart and liver of dead laying hens were collected. The growth characteristics and morphology of bacterial colonies were observed through isolation and culture of bacteria, smear microscopy, and biochemical identification. PCR technology and animal experiments were used to detect the amplification of 16S rRNA and the survival of infected animals for sequencing analysis and toxicity identification. Experiments of drug sensitivity on isolated bacterial strains were conducted to screen specific therapeutic drugs based on judgment criteria and guide schemes of clinical medication. Results The dead laying hens had obvious pathological features of pericarditis and hepatitis. The results of isolation, culture and identification of bacteria showed that the strain isolated was pathogenic E.coli. The case was diagnosed as infection of E.coli. The strain was completely resistant to 10 antibiotics including florfenicol and chloramphenicol, but highly sensitive to polymyxin B, ofloxacin, norfloxacin, piperacillin, ceftriaxone and cefotaxime. Conclusions The strain isolated had multi-drug resistance. The results of the drug sensitivity experiment can be used as a reference for clinical medication in this farm.
