PRRS NADC30 GX-R18弱毒株对后备母猪免疫驯化效果的评估
Evaluating the effects of PRRS NADC30 GX-R18 attenuated strain on the immune domestication of replacement gilts
目的 为评估PRRS NADC30 GX-R18弱毒株对后备母猪的驯化效果。 方法 选取某后备猪场1 930头PRRSV抗体阳性、抗原阴性的后备母猪,使用“两活一灭”的免疫驯化方式进行研究。分别在全场进满苗后的第7天和第28天免疫1针NADC30 GX-R18弱毒株、第58天免疫1针PRSS灭活疫苗。分别在免疫前、一免后21 d、二免后30 d、三免后30、45、60、90 d和转至母猪场后30 d进行前腔静脉采血,检测血液中PRRSV和N蛋白抗体。 结果 抗原单阴的断奶仔猪在免疫NADC30 GX-R18弱毒株后,分别在二免后30 d和三免后30 d检测到PRRSV病毒血症,其他阶段血液及引种前后备母猪扁桃体PRRSV检测呈阴性。一免后21 d和二免后30 d抗体显著上升,到三免后30 d达到100%,并持续到引种后30 d(N-ELISA抗体阳性率97.51%)。与免疫前相比,免疫后不同阶段抗体水平极显著升高(P<0.01),且抗体S/P值呈正态分布。生产性能统计分析该批次猪只生产成绩稳定。 结论 NADC30 GX-R18弱毒株具有高度安全性,对后备母猪免疫驯化效果良好,可以用于PRRS阳性后备母猪的免疫驯化。
Objectives The effects of porcine reproductive and respiratory syndrome virus (PRRSV) NADC30 GX-R18 attenuated strain on the immune domestication of replacement gilts were evaluated. Methods 1 930 PRRS antibody-positive but antigen-negative replacement gilts from a replacement pig farm were selected to study the immunization domestication with 'two-live-one-inactivated' immunization protocol. The replacement gilts were vaccinated with the NADC30 GX-R18 attenuated strain on day 7 and day 28 after the introduction of the vaccine to the farm, followed by a PRRS inactivated vaccine on day 58. Blood samples were collected from the anterior vena cava before immunization, and on day 21 after the first immunization, day 30 after the second immunization, day 30, 45, 60, and 90 after the third immunization, and day 30 after transfer to the pig farm and detected for antibodies against PRRSV pathogens and N-protein. Results PRRS viremia was detected in antigen-negative weaned piglets on day 30 after the second immunization and day 30 after the third immunization following vaccination with the PRRSV NADC30 GX-R18 attenuated strain. PRRSV was negative in blood samples at other stages and in gilt tonsils before introduction. Levels of antibody increased significantly on day 21 after the first immunization and day 30 after the second immunization, reaching 100% seropositivity by day 30 after the third immunization and persisting until 30 days after introduction (97.51% N-ELISA antibody positivity). Levels of antibody significantly increased at different stages after immunization (P<0.01) compared with that before immunization, with S/P values normally distributed. The results of statistically analyzing the production performance showed that the productivity in this batch of pigs was stable. Conclusions PRRS NADC30 GX-R18 attenuated strains have high safety and good effects of immune domestication on replacement gilts, and can be used for the immune domestication of replacement gilts after being PRRS positive.
