基于高分辨质谱的饲料中3种聚醚类药物的快速测定
Rapid determination of 3 polyether antibiotics in feed based on high-resolution mass spectrometry
目的 建立基于高分辨质谱快速测定方法,用于检测饲料中盐霉素、甲基盐霉素和莫能菌素,实现对3种聚醚类药物的定量分析,为饲料监管提供技术支持。 方法 饲料样品经乙腈-水溶液(含0.1%甲酸)提取离心去除沉淀后,上清液经Oasis PRiME HLB固相萃取柱净化,所得净化液利用超高效液相色谱-四级杆-静电场轨道阱高分辨质谱仪(UPLC-Q-Orbitrap HRMS)电喷雾正离子模式,Full MS/dd-MS2(全扫描/数据依赖型二级质谱)扫描模式开展数据采集,并对盐霉素、甲基盐霉素和莫能菌素进行定量分析。 结果 3种聚醚类药物在2~100 ng/mL范围内线性关系良好,相关系数R2≥0.991 0,饲料中3种聚醚类药物的平均回收率为92.9%~98.5%,相对标准偏差(RSD)为2.3%~3.5%。 结论 所建立的高分辨质谱方法具有较好的准确性和稳定性,适用于饲料中3种聚醚类药物的快速检测。
Objectives A method of rapidly determining 3 polyether antibiotics including salinomycin, narasin, and monensin in feed was established based on high-resolution mass spectrometry to quantitatively analyze them and provide technical support for feed regulation. Methods The samples of feed were extracted with acetonitrile-water solution (containing 0.1% formic acid) and centrifuged to remove the precipitate. The supernatant was purified via SPE using an Oasis PRiME HLB column. The purified solution obtained was used for collecting data with the UPLC-Q-Orbitrap HRMS electrospray positive ion mode and the Full MS/dd-MS2 (full scan/data dependent secondary acquisition) scanning mode. Quantitative analysis was conducted on salinomycin, narasin, and monensin. Results The linear relationship between the 3 polyether antibiotics was good within the concentration range of 2-100 ng/mL, with a correlation coefficient R2≥0.991 0. The average recovery rate of 3 polyether antibiotics in feed was 92.9%-98.5%, with a relative standard deviation (RSD) of 2.3%-3.5%. Conclusions The method with high-resolution mass spectrometry established has good accuracy and stability, and is suitable for rapid detection of 3 polyether antibiotics in feed.
