皂翘消癖颗粒通过调控ER/RAS-RAF-ERK通路干预肝郁痰凝型乳腺癌癌前病变的机制研究

皂翘消癖颗粒通过调控ER/RAS-RAF-ERK通路干预肝郁痰凝型乳腺癌癌前病变的机制研究
Mechanism of Zaoqiao Xiaopi Granules in intervening breast cancer precancerous lesions with Ganyu Tanning pattern via regulating the ER/RAS-RAF-ERK pathway
目的 构建SD大鼠肝郁痰凝型乳腺癌癌前病变模型,探讨皂翘消癖颗粒的作用及分子机制。 方法 40只SD雌性大鼠按体重随机分为空白对照组、模型Ⅰ组(DMBA+雌、孕激素诱导癌前病变)、模型Ⅱ组(模型I+6周应激诱导肝郁痰凝型乳腺癌癌前病变模型)和皂翘消癖组。皂翘消癖组于造模后给药干预。HE染色观察乳腺病理学改变,免疫组化检测ERα、RAS、ERK2和p-ERK表达水平。 结果 血清5-HT含量比较:实验第9周末,模型Ⅱ组与皂翘消癖组(肝郁痰凝模型)显著高于模型Ⅰ组,空白组最低;实验第13周末,模型Ⅱ组血清5-HT含量最高,其余各组由高至低依次为模型Ⅰ组、皂翘消癖组、空白组。乳腺癌癌前病变发生率:皂翘消癖组显著低于模型Ⅰ组和模型Ⅱ组(P<0.05);病变程度以模型Ⅱ组最重,模型Ⅰ组次之,皂翘消癖组最轻,且模型Ⅱ组出现1例原位癌。乳腺组织ERα、RAS、ERK2及p-ERK表达:模型Ⅰ组、模型Ⅱ组及皂翘消癖组均高于空白组(P<0.05),模型Ⅱ组高于模型Ⅰ组(P<0.05),皂翘消癖组低于模型Ⅰ组和模型Ⅱ组(P<0.05)。 结论 在DMBA诱导基础上联合雌、孕激素序贯刺激及慢性不可预见性应激,可成功地建立肝郁痰凝型乳腺癌癌前病变大鼠模型。皂翘消癖颗粒能显著降低该大鼠乳腺癌癌前病变的发生率及非典型增生程度,并抑制其恶性进展;其机制与下调乳腺组织中ERα、RAS、ERK2及p-ERK的表达,从而抑制RAS-RAF-ERK信号通路的异常激活有关。
Objective To establish a rat model of breast cancer precancerous lesions with the Ganyu Tanning (liver depression and phlegm coagulation) pattern and investigate the effects and underlying molecular mechanisms of Zaoqiao Xiaopi (ZQXP) Granules. Methods 40 female SD rats were randomly divided into four groups (n=10 per group): control group, model I group (induced with DMBA and sequential estrogen/progesterone stimulation), model II group (model I + 6 weeks of chronic unpredictable stress to establish the Ganyu Tanning pattern model) and the ZQXP group (Ganyu Tanning model treated with ZQXP Granules after modeling). Mammary gland pathology was observed via HE staining. The expression levels of ERα, RAS, ERK2, and p-ERK in mammary tissues were detected by immunohistochemistry. Results Serum 5-HT levels: At week 9, levels in the model II and ZQXP groups (both Ganyu Tanning models) were significantly higher than in the model I group, with the control group being the lowest. At week 13, the model II group exhibited the highest serum 5-HT content, followed by the model I group, ZQXP group and control group. Incidence and severity of precancerous lesions: The incidence in the ZQXP group was significantly lower than in the model I and model II groups (P<0.05). Lesion severity was greatest in model II, followed by Model I, and was mildest in the ZQXP group. Notably, one case of carcinoma in situ was observed in the model II group. 3. Expression of ERα, RAS, ERK2, and p-ERK: Expression was higher in the model I, model II, and ZQXP groups compared to the control group (P<0.05). The model II group showed higher expression than model I (P<0.05), while the ZQXP group showed lower expression than both model I and model II groups (P<0.05). Conclusion A rat model of breast cancer precancerous lesions with the Ganyu Tanning pattern was successfully established using DMBA induction combined with sequential estrogen/progesterone stimulation and chronic unpredictable stress. ZQXP Granules significantly reduced the incidence and severity of atypical hyperplasia in this model and inhibited malignant progression. The mechanism may involve downregulating the expression of ERα, RAS, ERK2, and p-ERK in mammary tissue, thereby suppressing the aberrant activation of the RAS-RAF-ERK signaling pathway.
