干旱胁迫对紫锥菊种子萌发及生理特性的影响

干旱胁迫对紫锥菊种子萌发及生理特性的影响
The Effect of Drought Stress on the Germination and Physiological Characteristics of Echinacea purpurea Seeds
为研究干旱胁迫对紫锥菊种子萌发的影响,以蒸馏水(CK)、φ(PEG-6000)=5%(T1)、10%(T2)、15%(T3)、20%(T4)溶液模拟不同渗透势的干旱胁迫,通过室内发芽试验,测定紫锥菊种子的发芽势、发芽率、发芽指数和幼苗的胚根长、胚芽长、鲜质量、丙二醛浓度以及保护酶(SOD、CAT、POD)活性等指标。结果表明:①T1对紫锥菊种子的萌发没有明显抑制效果,T2、T3、T4对种子的萌发均起到显著抑制作用,且φ(PEG-6000)越高抑制作用越强。②与CK相比,T1幼苗的胚根、胚芽长与CK的差异不显著,但鲜质量显著降低,差异达到显著,而T2、T3、T4的胚根长、胚芽长、鲜质量均显著降低,差异达到显著水平。③随着φ(PEG-6000)增大,幼苗丙二醛浓度逐渐积累,除T1外,其他处理与CK之间的差异显著;而CAT活性的变化在不同φ(PEG-6000)处理间的差异不显著;POD活性呈现先下降后上升的趋势,除T3与CK之间的差异显著外,其他处理与CK之间的差异不显著;SOD活性随φ(PEG-6000)的增大呈现增加的趋势,除T3处理与CK之间的差异显著外,其他处理与CK之间的差异不显著。综上所属,在T3处理的干旱胁迫下,紫锥菊种子的发芽势和发芽指数下降的幅度较大,但种子萌发率仍较高,达到82.0%,且在胁迫程度低于T3时,能够维持较高的体内抗氧化酶活性。φ(PEG-6000)=15%可以作为紫锥菊萌发期室内模拟干旱胁迫的适宜的体积分数。
To investigate the effect of drought stress on the seed germination of Echinacea purpurea, solutions of distilled water (CK), 5% of φ(PEG-6000) (T1), 10% of φ(PEG-6000) (T2), 15% of φ(PEG-6000) (T3), 20% of φ(PEG-6000) (T4) were prepared to simulate drought stress with different osmotic potentials. Through indoor germination tests, the germination potential, germination rate, germination index of seeds and the root length, shoot length, fresh weight of seedlings, malondialdehyde (MDA) concentration and the activities of protective enzymes (SOD, CAT, POD) were determined. The results showed that ① T1 treatment had no significant inhibitory effect on the germination of Echinacea purpurea seeds, whereas treatments with T2, T3, and T4 concentrations significantly inhibited seed germination, with stronger inhibition observed at higher concentrations. ② Compared with CK, there was no significant difference in the embryo root length and embryo length of Echinacea purpurea seedlings treated with T1, but the fresh weight showed a significant reduction. However, the embryo root length, embryo length, and fresh weight under T2, T3, and T4 treatments were significantly reduced compared to CK. ③ As the PEG-6000 concentration increased, malondialdehyde content gradually accumulated. Except for T1, significant differences were observed between treatments and CK, while changes in CAT activity levels were not significantly different among the various PEG concentration treatments. POD activity initially decreased and then increased, with no significant differences observed between the other treatments and the control, except for T3 which showed a significant difference. SOD activity increased with increasing stress concentration, with the exception of a significant difference between the T3 treatment and CK; no significant differences were observed for the other treatments. In summary, when the drought stress concentration is at T3, the germination potential and germination index of Echinacea purpurea seeds significantly decrease, but the seed germination rate remains high, reaching 82.0%. Furthermore, when the stress concentration is below T3, high levels of in vivo antioxidant enzyme activity are maintained, making it a suitable PEG concentration for simulating drought stress during the germination period of Echinacea purpurea.
