基于UPLC-Q-TOF-MS/MS鉴定温胆宁心颗粒的化学成分研究

基于UPLC-Q-TOF-MS/MS鉴定温胆宁心颗粒的化学成分研究
Study on the identification of chemical constituents of Wendan Ningxin Granules based on UPLC-Q-TOF-MS/MS
目的 采用多反应监测模式的超高效液相色谱-串联质谱联用技术(UPLC-Q-TOF-MS/MS)法对温胆宁心颗粒的化学成分进行快速识别和鉴定。 方法 以Agilent SB-C18(2.1 mm×100 mm,1.8 μm)为色谱柱,超纯水-0.1%甲酸(A)、乙腈-0.1%甲酸(B)为流动相进行梯度洗脱;柱温:40 ℃,流速:0.35 mL·min-1。质谱采用电喷雾离子源(ESI)正、负离子模式,扫描范围m/z 100~1 500,利用Analyst 1.6.3处理质谱采集数据,然后基于MWDB数据库、HIT数据库、BATMAN数据库及相关文献共分析。 结果 通过与对照品比对,同时依据相对分子量、保留时间和质谱特征碎片离子信息及物质成分特定靶标生物的活性水平(level)对化合物进行综合鉴定,其中在10味植物类中药中鉴定出136个化学成分,包括62个黄酮类、13个生物碱类、22个萜类、20个酚酸类、8个木脂素和香豆素类、1个醌类及10个其他类化合物,2味矿物类中药鉴定出13个化学成分,合并去重后得到温胆宁心颗粒的147个化学成分。 结论 构建的UPLC-Q-TOF-MS/MS能快速、准确、较全面地鉴定温胆宁心颗粒的化学成分,为其药效物质基础的深入阐释提供了重要依据。
Objective To rapidly identify and characterize the chemical constituents of Wendan Ningxin Granules (WDNX) using ultra-performance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF-MS/MS) in multiple reaction monitoring mode. Methods The separation was performed on an Agilent SB-C18 column (2.1 mm × 100 mm, 1.8 μm) with ultra-pure water containing 0.1% formic acid (A) and acetonitrile containing 0.1% formic acid (B) as the mobile phase for gradient elution. The column temperature was set at 40 ℃ , and the flow rate was 0.35 mL·min⁻¹. Mass spectrometry was performed using an electrospray ionization (ESI) source in both positive and negative ion modes, with a scan range of m/z 100-1,500. The data from mass spectrometry were processed using Analyst 1.6.3 software, followed by comprehensive analysis based on the MWDB, HIT, and BATMAN databases as well as relevant literature. Results Compounds were comprehensively identified by comparison with reference standards, and based on relative molecular weight, retention time(RT), characteristic fragment ions, and the activity levels of specific target organisms for the constituent components. A total of 136 chemical constituents were identified in the 10 herbal medicines, including 62 flavonoids, 13 alkaloids, 22 terpenoids, 20 phenolic acids, 8 lignans and coumarins, 1 quinone, and 10 other compounds. Additionally, 13 chemical constituents were identified in the 2 mineral-based traditional Chinese medicines. After merging and removing duplicates, 147 chemical constituents of WDNX were obtained. Conclusion The established UPLC-Q-TOF-MS/MS method can rapidly, accurately, and comprehensively identify the chemical constituents of WDNX, providing an important basis for the indepth elucidation of its pharmacodynamic material basis.
