中药调控TGF-β/Smads通路抗心肌纤维化的机制研究进展

中药调控TGF-β/Smads通路抗心肌纤维化的机制研究进展
Research progress on the mechanism of anti-myocardial fibrosis by regulating transforming growth factor-β/Smads pathway with traditional Chinese medicine
心肌纤维化是多种遗传性与获得性心血管病变的核心终末病理环节,其发生驱动心脏结构与功能恶化,并通过影响电生理稳态参与疾病进程,是重要的治疗干预靶标。转化生长因子-β(TGF-β)在该过程中扮演关键角色,通过激活Smad依赖性和非依赖性信号网络,协同调控下游效应分子,促进细胞外基质沉积与心肌重构。TGF-β/Smads信号通路作为心肌纤维化研究中的经典通路,近年来在中药药理研究中备受关注。且有相关研究揭示,中药可通过调控该通路及其上下游关联因子,干预纤维化进程,实现一定程度的病理逆转。本文系统综述中药通过调控TGF-β/Smads通路抗心肌纤维化的作用机制,从分子及通路层面阐释中药靶向治疗的潜在策略与科学内涵。
Myocardial fibrosis is the core terminal pathological link of a variety of hereditary and acquired cardiovascular diseases. Its occurrence drives the deterioration of cardiac structure and function, and participates in the disease process by affecting electrophysiological homeostasis. It is an important therapeutic intervention target. Transforming growth factor-β (TGF-β) plays a key role in this process by activating Smad-dependent and Smad-independent signaling networks, synergistically regulating downstream effector molecules to promote extracellular matrix deposition and myocardial remodeling. As a classical pathway in the study of myocardial fibrosis, TGF-β/Smads signaling pathway has attracted much attention in the pharmacological research of traditional Chinese medicine in recent years. Related studies have revealed that traditional Chinese medicine can intervene in the fibrosis process by regulating the pathway and its upstream and downstream related factors, achieving a certain degree of pathological reversal. This article systematically reviews the mechanism of traditional Chinese medicine in anti-myocardial fibrosis by regulating the TGF-β/Smads pathway, and explains the potential strategies and scientific connotation of traditional Chinese medicine targeted therapy from the molecular and pathway levels.
