摘要

青蛤(Cyclina sinensis)是我国沿海滩涂广泛分布的重要经济贝类。然而,养殖过程中频繁遭遇的高温与病原微生物感染等挑战,严重制约其生长与存活。为深入解析青蛤Caspase-3的分子特征,及其在高温胁迫与副溶血弧菌(Vibrio parahaemolyticus)感染下的响应机制,本研究采用生物信息学方法,对青蛤Caspase-3的核苷酸与氨基酸组成、二级及三级结构进行分析,并开展氨基酸序列比对与系统发育树构建;同时通过荧光定量PCR实验,验证Caspase-3在高温单一胁迫及高温-副溶血弧菌双重胁迫下的表达模式,初步揭示其在青蛤逆境适应中的作用。

Abstract

Cyclina sinensis is an important economic bivalve species widely distributed along the coastal mudflats of China.However,frequent challenges such as high temperatures and pathogen microbial infections during the breeding process have severely restricted its growth and survival.To deeply analyze the molecular characteristics of Caspase-3 in Cyclina sinensis and its response mechanisms under high temperatures stress and Vibrio parahaemolyticus infection,this study employed bioinformatics methods to analyze the nucleotide and amino acid composition,secondary and tertiary structures of CsCaspase-3,and conducted amino acid sequence comparisons and phylogenetic tree construction.Additionally,through fluorescence quantitative PCR experiments,the expression patterns of CsCaspase-3 under single high temperatures stress and dual high-temperature-Vibrio parahaemolyticus stress were verified,and its functional role in Cyclina sinensis adaptation to adverse conditions was preliminarily revealed.
