基于HPLC指纹图谱相似度分析和化学模式识别的西藏药用麻黄品种研究
Study on the Medicinal Varieties of Xizang Ephedrae Herba Based on HPLC Fingerprint Similarity Analysis and Chemical Pattern Recognition
目的 基于HPLC图谱相似度分析和化学模式识别对西藏药用麻黄品种进行研究。 方法 HPLC采用Phenomenex luna-C18色谱柱(4.6 mm×250 mm,5 μm);流动相A:磷酸(0.05%)-三乙胺(0.05%)水溶液,流动相B:乙腈,梯度洗脱;柱温30℃;流速1.0 mL/min;检测波长200 nm。选取各批次麻黄药材色谱图中的主要共有峰作为变量,运用SPSS 21.0 软件进行聚类分析和主成分分析对药材进行化学模式识别。 结果 12批西藏药用麻黄HPLC指纹图谱中有8个共有峰,相似度在0.546~0.922之间;12批样品聚类分为4大类,与主成分分析结果基本一致,表现出明显的按生境聚类的特征。 结论 该方法稳定可靠,可为西藏药用麻黄品种研究和质量控制提供参考。
Objective To research the medicinal varieties of Xizang Ephedrae Herba, HPLC fingerprint similarity analysis and chemical pattern recognition were carried out. Methods For HPLC, separation was carried out on a Phenomenex luna-C18 column(4.6 mm×250 mm,5 μm)with a mobile phase consisting of 0.05% phosphoric acid-0.05% triethylamine aqueous solution(A) and acetonitrile(B) in a linear gradient. The column temperature was maintained at 30℃, the flow rate was kept at 1.0 mL/min, and the detection wavelength was set at 200 nm. The main common peaks in the chromatogram of each batch of ephedra were selected as variables, and SPSS 21.0 was used for cluster analysis and principal component analysis to carry out chemical pattern recognition for medicinal herbs. Results Eight common peaks were identified in 12 batches of Xizang Ephedrae Herba, with similarities ranging from 0.546 and 0.922; The 12 batches of samples were clustered into 4 categories, which was basically consistent with the results of principal component analysis, and showed obvious characteristics of clustering by habitat. Conclusion HPLC fingerprint similarity analysis combined with chemical pattern recognition could be applied to the medicinal variety identification and quality control of Xizang Ephedrae Herba due to its dependability and stability.
