摘要
目的 研究藏药坎巴的主要基原大籽蒿治疗藏医真布病的潜在作用机制。方法 通过蒸馏水提取结合甲醇萃取藏药大籽蒿的粗粉, 利用超高效液相色谱Q静电场轨道阱质谱联用技术(UPLC-Q-Orbitrap MS)进行定性鉴别; 鉴定的化合物利用SwissTargetPrediction等数据库筛选出关键成分及其化合物靶点, 利用KEGG通路数据库获取真布病靶点, 两者的交集靶点采用AutoDock等软件进行分子对接验证。结果 UPLC-Q-Orbitrap MS鉴定的249个化合物, 经-6 cm/s≤皮肤渗透系数(log Kp)≤-2 cm/s获得79个关键成分; 905个化合物靶点与95个真布病通路的靶点共有15个交集靶点; 分子对接中基质金属蛋白酶3(MMP3)与香叶木素、芹菜素、异泽兰黄素、药根碱、黄藤素等关键成分有强结合性, 结合能均≤-33.5 kJ/mol。结论 藏药五味甘露组方药材大籽蒿因疗效良好, 成为藏医临床常见治疗真布病的外治药物, 主要通过“黄水关节”路径治疗真布病, 其机制涉及抗炎、免疫调节、血管抑制、基质保护及破骨调控等作用, 可抑制MMP3活性并调控寒热型黄水病理, 从分子层面验证藏医“调黄水利关节”理论, 为《四部医典》黄水病机理论提供科学依据。
Abstract
Objective To study the potential mechanism of action of Artemisia sieversiana, the main substrate of the Tibetan medicine Kamba, in the treatment of Zhenbu disease in Tibetan medicine. Methods The crude powder of the Tibetan medicine Artemisia sieversiana was extracted by distilled water extraction combined with methanol, and qualitatively identified using ultra-performance liquid chromatography-quadrupole-Orbitrap mass spectrometry (UPLC-Q-Orbitrap MS). The identified compounds were screened for key components and their compound targets using SwissTargetPrediction and other databases, and the KEGG pathway database was utilized to obtain the targets of Zhenbu disease. The intersection targets of the two was verified by molecular docking using AutoDock and other software. Results Among the 249 compounds identified by UPLC-Q-Orbitrap MS, 79 key components were obtained by -6 cm/s≤skin permeability coefficient (Log Kp) ≤-2 cm/s. There were 15 intersection targets among 905 compound targets and 95 targets of the Zhenbu disease pathway. In molecular docking, matrix metalloproteinase 3 (MMP3) had strong binding properties with key components such as diosmetin, apigenin, eupatilin, jatrorrhizine, and palmatine, and the binding energies were all ≤-33.5 kJ/mol. Conclusion The Tibetan medicine Artemisia sieversiana has become a common external treatment for Zhenbu disease in Tibetan medicine because of its good therapeutic effect, and it mainly treats Zhenbu disease through the pathway of “yellow water joints”, whose mechanism involves anti-inflammation, immune regulation, vascular inhibition, matrix protection and osteoclastic regulation, etc. It can inhibit MMP3 activity and regulate the pathology of cold-heat type yellow water, which verifies the theory of Tibetan medicine of “eliminating yellow water and facilitating joints” at the molecular level, and provides a scientific basis for the theory of yellow water pathology in the “Four Medical Classics”.
