诺如病毒快速免疫层析检测效能的评估及应用
Evaluation of the detection performance and application for rapid immunochromatographic test of norovirus
目的 评估一种诺如病毒免疫层析(Immunochromatography,IC)检测试剂盒的检测效能,为免疫层析法在实际筛查检测中的应用提供依据。 方法 收集100份腹泻样本,同时采用IC检测试剂盒与诺如病毒检测中推荐的实时定量逆转录聚合酶链式反应(Real-time quantitative reverse transcription PCR,RT-qPCR)方法对样本平行检测,通过对2种方法检测结果进行统计分析,评估IC检测试剂盒对诺如病毒及其基因型的检测效能。 结果 IC检测试剂盒检测诺如病毒的灵敏度、特异性和符合率分别达到了95.9%、100.0%和97.0%。IC检测试剂盒与RT-qPCR检测结果比较=1.333,P=0.248,差异无统计学意义。进一步对不同诺如病毒基因型进行检测,结果显示,IC检测试剂盒对诺如病毒GⅠ.5、GⅡ.3和GⅡ.7基因型检测灵敏度均为100.0%,而对GⅡ.4和GⅡ.17基因型检测灵敏度分别为94.4%和87.5%。 结论 IC检测试剂盒检测诺如病毒具有较高的灵敏度和特异性,检测结果与RT-qPCR检测的一致性高,可作为基层医疗机构诺如病毒筛检方法。
Objective : To evaluate detection performance of a norovirus immunochromatographic (IC) test kit and provide evidence for its practical application. Methods A total of 100 diarrheal samples were collected and tested in parallel using the IC test kit and the RT-qPCR methods recommended by quarantine industry standards. The detection efficacy of the IC test kit for norovirus and its genotyping was assessed through statistical analysis of results from both methods. Results The sensitivity, specificity, and concordance rate of the IC detection kit reached 96%, 100%, and 97%, respectively. Compared with the standard detection methods, the McNemar chi-square value was 1.333, and the P-value was 0.248, indicating no statistical difference. The results of further detection of different norovirus genotypes showed that the IC detection kits demonstrated a sensitivity of 100.0% for the norovirus GⅠ.5, GⅡ.3, and GⅡ.7 genotypes, while the sensitivities for the GⅡ.4 and GⅡ.17 genotypes were 94.4% and 87.5%, respectively. Conclusion The IC detection kit has high sensitivity and specificity, the detection results show high consistency with RT-qPCR detection, making it suitable as a screening method for norovirus in primary healthcare institutions.
