The Mechanistic Role of Chemerin in the Impact of Aerobic Exercise on Mouse Appetite

摘要

为了探究chemerin在有氧运动影响小鼠食欲中的作用及机制。利用8周龄野生型（WT）、脂肪chemerin敲除[chemerin(-/-)]小鼠完成6周饮食+6周的有氧运动干预。干预结束后，禁食12h，取材。利用ELISA方法检测实验小鼠血清chemerin、leptin水平；qRT-PCR方法检测动物下丘脑组织中chemerin、leptin、NPY、AgRP、POMC和CART等食欲调节因子mRNA表达水平。结果显示，脂肪chemerin敲除均可降低小鼠摄食量，但有氧运动并没有进一步增强chemerin敲除使小鼠摄食量降低的效应。有氧运动和脂肪chemerin敲除均可降低血清chemerin、leptin水平和小丘脑中食欲促进因子AgRP的mRNA表达水平，并使下丘脑中leptin和食欲抑制因子POMC和CART的mRNA表达水平升高，且未见有氧运动和chemerin敲除的叠加效应。提示，chemerin减少在有氧运动降低小鼠食欲中发挥重要作用，且该作用可能与其上调下丘脑leptin的mRNA表达水平、调控下丘脑食欲调节因子的mRNA表达水平有关。

Abstract

To investigate the role and mechanism of chemerin in the effect of aerobic exercise on mouse appetite, 8-week-old wild-type (WT) and adipose chemerin knockout [chemerin (-/-)] mice were subjected to a 6-week dietary intervention followed by 6 weeks of aerobic exercise training. After the intervention, fasting for 12 hours was performed before sample collection. Serum chemerin and leptin levels were measured using ELISA, while mRNA expression levels of appetite-regulating factors—including chemerin, leptin, NPY, AgRP, POMC, and CART—in hypothalamic tissues were analyzed by qRT-PCR. The results demonstrated that adipose chemerin knockout significantly reduced food intake in mice, whereas aerobic exercise did not further enhance this reduction. Both aerobic exercise and adipose chemerin knockout decreased serum chemerin and leptin levels as well as the mRNA expression of the appetite-promoting factor AgRP in the hypothalamus, while increasing the mRNA expression levels of leptin and the appetite-suppressing factors POMC and CART. No synergistic effect was observed between aerobic exercise and chemerin knockout. These findings suggest that reduced chemerin levels play a critical role in the appetite-reducing effect of aerobic exercise in mice, likely through upregulation of hypothalamic leptin mRNA expression and modulation of hypothalamic appetite-regulating factor mRNA levels.
