摘要

以桑叶提取物总黄酮树脂解吸率为指标,通过单因素试验和响应面分析法对AB-8大孔树脂不同再生工艺解吸桑叶提取物的工艺条件进行对比研究。考察了AB-8大孔树脂在不同再生清洗试剂浓度、树脂再生清洗水流速、树脂再生清洗水温等再生工艺下,对桑叶提取物总黄酮解吸率的影响,采用Box-Behnken试验设计方法,对影响桑叶提取物总黄酮解吸率的各因素进行显著性及交互作用分析,最终确认AB-8大孔树脂解吸桑叶提取物总黄酮的最适宜工艺条件。AB-8大孔树脂解吸桑叶提取物总黄酮的最佳再生工艺条件为:树脂再生清洗试剂质量分数为9%、树脂再生清洗水流速为2 BV/h、树脂再生清洗水温度为50 ℃。经实验探索和中试验证,该工艺具有较好的重现性。因此,优化AB-8大孔树脂的再生工艺可提升解吸桑叶提取物总黄酮的稳定性。研究结果表明,AB-8大孔树脂的不同再生工艺对桑叶提取物总黄酮解吸率影响显著。

Abstract

The desorption rate of total flavonoid resin of mulberry leaf extract was indicated,the process conditions of mulberry leaf extract applied in different regeneration processes of AB-8 macroporous resin were compared by single factor test and response surface analysis.The regeneration processes of AB-8 macroporous resin under different regeneration processes, such as the concentration of regeneration cleaning reagent, the flow rate of resin regeneration cleaning water, and the water temperature of resin regeneration cleaning, were investigated,effect on the desorption rate of total flavonoids in mulberry leaf extract,the Box-Benhnken experimental design method was used,the significance and interaction of the factors affecting the desorption rate of total flavonoids in mulberry leaf extract were analyzed,finally, it was confirmed that the AB-8 macroporous resin regeneration process had the most suitable process conditions for the desorption rate of total flavonoids in mulberry leaf extract.The optimal process conditions for AB-8 macroporous resin regeneration were as follows: resin regeneration cleaning reagent concentration of 9%, resin regeneration cleaning water flow rate of 2 BV/h, and resin regeneration cleaning water temperature of 50 ℃.After experimental exploration and pilot verification, the related process has good reproducibility.Therefore,The regeneration process conditions of AB-8 macroporous resin could improve the stability of the desorption rate of total flavonoids in mulberry leaf extract.The results of the study showed that different regeneration processes of AB-8 macroporous resin had significant effects on the desorption rate of total flavonoids in mulberry leaf extract。
