顶空气相色谱法测定艾曲波帕原料药中残留溶剂
Determination of residual solvents in Eltrombopag Olamine active pharmaceutical ingredients by headspace gas chromatography
目的 建立同时测定艾曲波帕原料药中8种有机溶剂(甲醇、乙醇、二氯甲烷、乙酸乙酯、四氢呋喃、三乙胺、正庚烷和吡啶)残留量的顶空气相色谱法。 方法 色谱柱为DB-1毛细管色谱柱(30 m×0.53 mm×2.65 μm),程序升温,进样口温度为250 ℃,检测器为火焰离子化检测器,温度为250 ℃,载气为高纯氮气,流速为2.5 mL/min,分流比为10∶1,顶空瓶平衡温度为90 ℃,平衡时间为25 min。对建立的方法进行专属性、线性和范围、检测限和定量限、精密度、重复性、加样回收率、稳定性考察。 结果 艾曲波帕原料药中8种残留溶剂(甲醇、乙醇、二氯甲烷、乙酸乙酯、四氢呋喃、三乙胺、正庚烷和吡啶)在一定浓度范围内显示良好的线性(r>0.999),定量限分别为6.0,7.6,6.2,4.0,1.5,0.76,0.75,3.9 μg/mL;检测限分别为2.0,2.5,2.1,1.3,0.5,0.25,0.25,1.3 μg/mL;平均回收率分别为102.4%,101.7%,101.6%,100.6%,101.4%,98.2%,100.7%和99.9%;RSD值分别为1.9%,1.7%,1.9%,1.7%,2.1%,1.9%,1.5%和1.5%(n=9);专属性、精密度、重复性、稳定性均符合方法学验证要求。 结论 本研究所建立的顶空气相色谱法适用于艾曲波帕原料药中8种残留溶剂的测定。
Objective To establish a headspace gas chromatographic method for the simultaneous determination of eight residual organic solvents(methanol, ethanol,dichloromethane, ethyl acetate, tetrahydrofuran, triethylamine, heptane and pyridine) in Eltrombopag Olamine active pharmaceutical ingredients(API). Methods The chromatographic column was DB-1 capillary column (30 m×0.53 mm×2.65 μm) with temperature-programmed, the temperature of the injection port was 250 ℃, the detector was flame ionization detector(FID) with the temperature of 250 ℃, the carrier gas was high purity nitrogen gas with the flow rate of 2.5 mL/min, the split ratio was 10∶1, the headspace heating temperature was 90 ℃, and the equilibrium time was 25 min. Specificity, linearity and range, detection limit and quantification limit, precision, repeatability, recovery, stability of the method were validated. Results Each of the eight residual solvents (methanol, ethanol, dichloromethane, ethyl acetate, tetrahydrofuran, triethylamine, heptane and pyridine) had good linearity within a certain concentration range(r>0.999), the limits of quantitation(LOQ) were 6.0, 7.6, 6.2, 4.0, 1.5, 0.76, 0.75, 3.9 μg/mL, and the limits of detection(LOD) were 2.0, 2.5, 2.1, 1.3, 0.5, 0.25, 0.25, 1.3 μg/mL. The average recoveries were 102.4%, 101.7%, 101.6%, 100.6%, 101.4%, 98.2%, 100.7% and 99.9%, and the RSD values were 1.9%, 1.7%, 1.9%, 1.7%, 2.1%, 1.9%, 1.5% and 1.5%(n=9), respectively. The specificity, precision, repeatability, and stability met the requirements of method validation. Conclusion The established headspace gas chromatographic method is suitable for the simultaneous determination of eight residual solvents in eltrombopag olamine API.
