ICP⁃MS法测定羊胎盘中多种微量元素含量
Determination of multiple trace elements in sheep placenta by ICP-MS
目的 建立羊胎盘中多种微量元素电感耦合等离子体质谱法(ICP-MS)含量测定的方法。 方法 使用微波消解法对羊胎盘样品粉末进行前处理,以锗、铟、铋、钪作为内标物进行校正,采用标准曲线法对微量元素进行含量测定并进行方法学研究。 结果 锰元素线性方程为y=92 885.195 0x+11 045.774 3(n=5,r=0.999 5);铝元素线性方程为y=38 950.504 8x+1 892.531 2(n=5,r=0.995 4);铜元素线性方程为y=33 111.860 7x+3 165.130 6(n=5,r=0.999 5);锌元素线性方程为y=10 273.228 4x+8 381.907 7(n=5,r=0.999 4);硒元素线性方程为y=624.307 3x+342.218 7(n=5,r=0.997 8);钴元素线性方程为y=67 175.235 8x+257.846 1(n=5,r=0.999 8);钼元素线性方程为y=6 495.782 4x+65.225 9(n=5,r=0.997 5);铅元素线性方程为y=196.925 5x+3.447 6(n=5,r=0.998 6);砷元素线性方程为y=8 066.305 5x+119.404 7(n=5,r=0.999 7);镉元素线性方程为y=22 460.203 8x+45.362 6(n=5,r=0.999 2);汞元素线性方程为y=64.446 8x+1.885 5(n=5,r=0.996 9)。检测方法精密度的相对标准偏差(RSD)<3.0%;重复性的相对标准偏差(RSD)<4.0%;加标回收率为92.59%~108.33%。 结论 本法简单易行,可操作性强,可用于羊胎盘微量元素的含量测定。
Objective To establish a method for determining the contents of multiple trace elements in sheep placenta by inductively coupled plasma mass spectrometry(ICP-MS). Methods Microwave digestion was used to prepare powders of sheep placenta samples. Ge,In,Bi and Sc were used as internal standards for calibration.The standard calibration curve method was used to determine the contents of trace elements and carry out methodological research. Results The linear equations were y=92 885.195 0x+11 045.774 3 for Mn(n=5,r=0.999 5), y=38 950.504 8x+1 892.531 2 for Al(n=5,r=0.995 4), y=33 111.860 7x+3 165.130 6 for Cu(n=5,r=0.999 5), y=10 273.228 4x+8 381.907 7 for Zn(n=5,r=0.999 4), y=624.307 3x+342.218 7 for Se(n=5,r=0.997 8), y=67 175.235 8x+257.846 1 for Co(n=5,r=0.999 8), y=6 495.782 4x+65.225 9 for Mo(n=5,r=0.997 5), y=196.925 5x+3.447 6 for Pb(n=5,r=0.998 6), y=8 066.305 5x+119.404 7 for As(n=5,r=0.999 7), y=22 460.203 8x+45.362 6 for Cd(n=5,r=0.999 2), y=64.446 8x+1.885 5 for Hg(n=5,r=0.996 9). The RSD of precision was less than 3.0%, the RSD of repeatability was less than 4.0%, and the recoveries were 92.59%-108.33%. Conclusion The method is simple and feasible, and it can be used for the determination of trace elements in sheep placenta.
