复方柏芪丸提取工艺优化及其HPLC指纹图谱建立

Optimization of extraction process and establishment of HPLC fingerprint of Compound Boqi pills
目的 优化复方柏芪丸的提取工艺并建立HPLC指纹图谱,为该制剂质量标准化提供依据。 方法 基于文献研究和前期吸水率实验结果,以加水量、提取时间、提取次数为考察因素,以地黄苷D、盐酸小檗碱、盐酸黄柏碱、金丝桃苷含量及干膏得率的综合得分为评价指标,采用等权重综合评分体系,通过三因素三水平正交实验,优化复方柏芪丸的提取工艺;对9批样品建立复方柏芪丸指纹图谱,结合聚类分析和偏最小二乘判别分析(PLS-DA)法进行化学模式识别,筛选复方柏芪丸的质量标志物。 结果 结合传统临床用药方式,本研究采用水煎煮法进行提取。经正交实验筛选,复方柏芪丸的最优提取工艺为加水量12倍,煎煮60 min,煎煮3次,验证试验综合得分平均值为98.73,工艺稳定可行。9批复方柏芪丸样品HPLC指纹图谱相似度均大于0.93,标定13个共有峰,通过聚类分析将9批样品分为3类,指认出地黄甘D、盐酸黄柏碱、盐酸小檗碱、金丝桃苷4种成分。 结论 该提取工艺稳定高效,建立的HPLC方法灵敏、简便、准确,可用于复方柏芪丸质量控制。
Objective To optimize the extraction process of Compound Boqi pills and establish an HPLC fingerprint profile, providing a basis for the standardization of this preparation. Methods Based on literature and preliminary water absorption rate experiments, water addition, extraction time, and extraction cycles were chosen as investigated factors. With the comprehensive score of rehmannioside D, berberine hydrochloride, oxycholine hydrochloride, and hyperoside contents and the dry extract yield as the evaluation index, an equal-weighted comprehensive scoring system was adopted, and the extraction process of Compound Boqi pills was optimized through a three-factor, three-level orthogonal experiment. HPLC fingerprints were established for nine batches of samples, and chemical pattern recognition was performed through cluster analysis and partial least squares discriminant analysis(PLS-DA) to screen quality markers for Compound Boqi pills. Results Based on traditional clinical administration methods, the decoction extraction was employed. Through orthogonal experiments, the optimal extraction process for Compound Boqi pill was determined to be: 12-fold water volume, decocting for 60 min, and three decoction times. The average comprehensive score of validation tests was 98.73, indicating stable and feasible process conditions. The HPLC fingerprint similarity of nine batches of Compound Boqi pill samples was all greater than 0.93, with 13 common peaks identified. Cluster analysis classified the nine batches into three groups, and identified four components: rehmannioside D, oxycholine hydrochloride, berberine hydrochloride, and hyperoside. Conclusion The extraction process is stable and efficient. The established HPLC method is sensitive, simple, and accurate, and can be used for the quality control of Compound Boqi pill.
