兔干扰素α和白介素2重组融合蛋白菌株构建与蛋白表达
Construction and protein expression of the recombinant fusion protein strains of rabbit interferon-α and interleukin-2
目的 本试验拟表达外源兔白细胞介素2(Interleukin-2, IL-2)和干扰素α(Interferon-alpha, IFN-α)重组蛋白,为兔病毒性疾病防治提供科学依据。 方法 参考GenBank数据库中的IL-2和IFN-α基因序列,去除信号肽后使用linker连接成为嵌合基因,将其克隆至原核表达载体pET-32a中并进行密码子优化,转化至E.coli BL21感受态细胞培养并诱导表达,通过SDS-PAGE对表达产物进行分析。 结果 pET32a-RaIFNα-IL2在约1 073 bp处有明显条带,表明质粒构建成功。pET32a-RaIFNα-IL2-BL21菌株在扩大培养3.5 h,1.0 mM IPTG诱导4 h后表达量较高,相对分子质量约为49.1 ku,主要以包涵体形式存在。 结论 成功构建了兔IL-2和IFN-α的重组蛋白。
Objectives This experiment aimed to express recombinant proteins of exogenous rabbit interleukin-2 (IL-2) and interferon-alpha (IFN-α) to provide a scientific basis for the clinical prevention and treatment of viral diseases in rabbit. Methods The gene sequences of rabbit IL-2 (accession number: NM_001163180.1) and IFN-α (accession number: XM_002708064.2) in the GenBank database were referred to remove the signal peptide and link to form a chimeric gene with a linker. The chimeric gene was cloned into the prokaryotic expression vector pET-32a with the codon being optimized, and then transformed into E. coli BL21 competent cell, cultured to induce expression. SDS-PAGE was used to analyze the expression product. Results pET32a-RaIFNα-IL2 had a distinct band at approximately 1 073 bp, indicating that the construction of recombinant plasmid was successful. The pET32a-RaIFNα-IL2-BL21 strain had a high level of expression after 3.5 h of expanded culture and 4 h of induction with 1.0 mM IPTG, with a relative molecular weight of approximately 49.1 ku, mainly in the form of inclusion bodies. Conclusions The construction of rabbit IL-2 and IFN-α recombinant proteins was successful.
