美洲大蠊多肽提取工艺优化及免疫活性验证
Optimization of Periplaneta americana polypeptide extraction process and verification of immune activity for Periplaneta americana
为探究美洲大蠊多肽的最佳提取工艺并对其免疫活性进行评价,以美洲大蠊为原料,通过对比超声波细胞破碎、胰蛋白酶酶解、中性蛋白酶酶解3种不同的提取方法,发现胰蛋白酶酶解提取效率最高.进一步探究酶底比、料液比、超声时间对美洲大蠊多肽得率的影响;采用CCK - 8法和中性红吞噬实验检测美洲大蠊多肽对RAW264.7细胞增殖和吞噬活性的影响.结果表明在酶底比5 000 U/g、料液比1∶20 g/mL、超声时间5 h的工艺条件下美洲大蠊多肽得率较高,为(11.92 ± 0.31)%.CCK - 8及中性红吞噬实验结果表明以质量浓度5 ~ 40 μg/mL给药时,RAW264.7细胞具有较好的增殖以及吞噬活力.体内实验结果亦表明美洲大蠊多肽在小鼠体内表现出较好的免疫活性.
In order to explore the optimum extraction process of Periplaneta americana polypeptide and evaluate its immune activity, three different extraction methods of ultrasonic cell disruption, trypsin digestion and neutral protease digestion were compared by using Periplaneta americana as the raw material. It was found that trypsin digestion had the highest extraction efficiency. The effects of enzyme-to - substrate ratio, solid-liquid ratio and ultrasonic time on the yield of Periplaneta americana polypeptide were further investigated. The effects of Periplaneta americana polypeptide on the proliferation and phagocytic activity of RAW264.7 cells were detected by CCK - 8 method and neutral red phagocytosis assay. The results showed that the yield of Periplaneta americana polypeptide was (11.92 ± 0.31)% under the conditions of enzyme - substrate ratio 5 000 U/g, solid-liquid ratio 1∶20 g/mL and ultrasonic time 5 h. The results of CCK - 8 and neutral red phagocytosis experiments showed that RAW264.7 cells had better proliferation and phagocytic activity when the mass concentration was 5 to 40 μg/mL. The results of in vivo experiments also showed that Periplaneta americana polypeptide showed good immune activity in mice.
