锅铲叶总黄酮提取工艺及体外抗氧化活性研究

锅铲叶总黄酮提取工艺及体外抗氧化活性研究
Study on extraction technology and antioxidant activity in vitro of total flavonoids from Passiflora wilsonii Hemsl foliage
以锅铲叶为原料,以乙醇体积分数、液料比、提取时间和提取温度为自变量,黄酮得率为因变量,通过单因素实验和响应面分析法,优化锅铲叶总黄酮的乙醇回流提取工艺.此外,以维生素C和芦丁为阳性对照,通过1,1 - 二苯基 - 2 - 三硝基苯肼(DPPH)、2,2' - 联氮 - 双 - 3 - 乙基苯并噻唑啉 - 6 - 磺酸(ABTS)和铁离子还原/抗氧化能力法(FRAP) 3种方法评价锅铲叶总黄酮的抗氧化能力.结果表明液料比、提取温度和时间会显著影响乙醇回流提取锅铲叶总黄酮的得率,以体积分数为70%的乙醇溶液为溶剂,在液料比为27 mL/g,61 ℃条件下回流提取42 min,黄酮平均得率可达23.4485 mg/g.提取所得总黄酮对DPPH、ABTS+和Fe3+ - TPTZ等均具有较好的清除能力,表现出良好的抗氧化活性,为进一步解析锅铲叶化学成分和活性研究提供重要的研究基础.
Taking Passiflora wilsonii Hemsl foliage as raw material and based on single factor experiment, the effects of ethanol volume fraction, liquid - solid ratio, extraction time and extraction temperature on the yield of total flavonoids from P. wilsonii Hemsl foliage were investigated. The antioxidant capacity of total flavonoids from P. wilsonii Hemsl was evaluated by DPPH, ABTS and FRAP, vitamin C and rutin were used as positive controls. The results showed that the yield of total flavonoids was significantly affected by liquid - solid ratio, extraction temperature and extraction time. With 70% ethanol solution as solvent, liquid - solid ratio of 27 mL/g, and extraction at 61 ℃ for 42 min, the average yield of flavonoids was 23.448 5 mg/g. The extracted total flavonoids had good scavenging ability for DPPH, ABTS+ and Fe3+ - TPTZ, providing an valuable research foundation for further analysis of the chemical composition and activity of P. wilsonii Hemsl foliage.
