野生贝壳状革耳的生物学特性及驯化栽培
Biological Characteristics and Domestic Cultivation of Panus conchatus
对采自西藏农牧学院校园柳树上的一株野生食用菌进行分离,编号为PC01。根据形态学特征和ITS序列聚类分析鉴定为贝壳状革耳(Panus conchatus)。为了充分开发和利用该种食用菌资源,对该种的生物学特性和驯化栽培进行了研究。结果表明:贝壳状革耳菌丝生长最佳碳源为麦芽糖,最佳氮源为酵母膏,最适pH为5.0,最适温度为25 ℃,最佳无机盐为K2HPO4。正交试验表明,贝壳状革耳菌丝生长的最佳配方为:为麦芽糖20.0 g·L-1,酵母膏3.0 g·L-1,pH为6.0,磷酸二氢钾2.5 g·L-1。驯化结果表明,以棉籽壳为主要栽培料可获得子实体,而以木屑为栽培主料可使贝壳状革耳菌丝在 30 d 左右满瓶,但无法获得子实体。
A wild edible mushroom strain PC01 was isolated from willow trees on the campus of Tibet Agricultural and Animal Husbandry University. Based on morphological features and ITS sequence clustering analysis, it was identified as a shell like fungus (Panus conchatus). In order to fully develop and utilize the edible mushroom resources, the biological characteristics and domestication cultivation of this species were studied. The results showed that the optimal carbon source for the growth of shell like fungus hyphae was maltose, and the optimal nitrogen source was yeast extract with the optimal pH 5.0, the optimal temperature 25℃ and the optimal inorganic salt K2HPO4. The orthogonal experiment showed that the optimal formula for the growth of shell shaped fungus hyphae was maltose 20.0 g·L-1, yeast extract 3.0 g·L-1, pH 6.0, and potassium dihydrogen phosphate 2.5 g·L-1. The domestication results showed that using cottonseed husks as the main cultivation material could obtain fruiting bodies, while using sawdust as the main cultivation material could make the shell like fungus hyphae fill the bottle in about 30 days, but did not obtain fruiting bodies.
