禽沙门氏菌、大肠杆菌及产气荚膜梭菌多重荧光PCR检测方法的建立及应用

禽沙门氏菌、大肠杆菌及产气荚膜梭菌多重荧光PCR检测方法的建立及应用
Establishment and Application of Multiplex Real-Time PCR Method for Avian Salmonella, Escherichia Coli and Clostridium Perfringens
为建立禽沙门氏菌、大肠杆菌及产气荚膜梭菌的多重荧光PCR鉴别检测方法,根据GenBank中沙门氏菌invA、大肠杆菌phoA、产气荚膜梭菌plc基因序列分别设计特异性引物和Taqman探针,并优化PCR反应体系和反应条件,应用该方法对临床样品进行检测。结果显示:该方法与巴氏杆菌、新城疫病毒和鸭疫里默氏杆菌等无交叉反应,具有较高特异性;沙门氏菌、大肠杆菌和产气荚膜梭菌最低检测限度(LOD)均达66.9 copies/μL,相比检测沙门氏菌、大肠杆菌及产气荚膜梭菌的普通PCR的敏感性分别高100倍、1 000倍和1 000倍,具有高敏感性;组内和组间变异系数均低于3.84%,稳定性良好;通过对30份临床样品和已鉴定的23份菌株进行检测,显示该检测方法符合率为100%。建立的多重荧光PCR方法特异性和敏感性较好,适用于临床禽沙门氏菌、大肠杆菌和产气荚膜梭菌单一或混合感染的快速诊断及流行病学调查,提高了家禽腹泻疾病的诊断效率,具有良好应用价值。
In order to establish a multiplex real-time PCR method for the differential detection of avian Salmonella, Escherichia coli and Clostridium perfringens. Specific primers and Taqman probes for Salmonella, Escherichia coli and Clostridium perfringens were designed based on invA, phoA and plc genes in GenBank, respectively, the PCR reaction system and reaction conditions were optimized. The clinical samples were detected by this method. The results showed that the method had high specificity and no cross-reaction with Pasteurella, Newcastle disease virus and Riemerella anatipestifer. The limit of detection (LOD) of Salmonella, Escherichia coli and Clostridium perfringens were all 66.9 copies/μL, 100 times, 1000 times and 1000 times more sensitive than general PCR for detection of Salmonella, Escherichia coli and Clostridium perfringens, indicating high sensitivity. Intra- and inter-assay coefficients of variation (CVs) of the Ct values were less than 3.84 %, indicating good stability. Through the detection of 30 clinical samples and 23 identified strains, the coincidence rate of the detection method was 100%. In summary, the established multiplex real-time PCR method demonstrated excellent specificity, sensitivity and detection efficiency. It was suitable for the rapid diagnosis and epidemiological investigation of investigation of avian Salmonella, Escherichia coli and Clostridium perfringens infections, either individually or in mixed infections. This method could enhance the efficiency of diagnosing avian diarrhea diseases and hold significant practical value.
