一株青稞根际溶磷真菌的鉴定及其促生特性研究
Identification of an Inter-Rhizosphate Solubilizing Fungus of Barley and Its Probiotic Properties
本研究从青稞根际土壤中筛选出一株溶磷真菌,并研究其促生特性,为提高青稞产量提供优质真菌菌剂。采用梯度稀释分离法,利用PDA、RBM、NBRIP固体培养基培养,从健康青稞根际土壤中分离筛选溶磷真菌,通过形态学和ITS序列分析对其进行鉴定;采用NBRIP液体培养测定菌株对Ca3(PO4)2的溶解能力;通过盆栽试验测定其对青稞的促生能力。 结果 (1)筛选出1株溶磷真菌菌株W41,将W41初步鉴定为毛茛葡萄孢盘菌(Botryotinia ranunculi);(2)在NBRIP液体培养基中有效磷含量为55.82 mg/L,并兼具固氮、解钾、产IAA的能力;(3)通过皿内发芽试验得到孢子液浓度为1×103 cfu/mL时青稞种子萌发生长效果最好;(4)盆栽试验中与对照相比,株高平均增加29.71%,根长平均增加28.69%,鲜重平均增加48.49%,干重平均增加39.12%,对青稞幼苗的促生效果显著。通过本研究,菌株W41具有较强的溶磷能力和促生效果,为利用真菌菌剂提高青稞产量提供了新的思路和方向。
In this study, a phosphorus solubilizing fungus was isolated from the rhizosphere soil of barley plants, and its growth-promoting characteristics were investigated. The aim was to provide a high-quality fungal agent to enhance barley yields. The isolation process involved a gradient dilution method, and the fungus was cultivated on solid media, including PDA, RBM, and NBRIP, using healthy barley rhizosphere soil samples. The fungus's identification relied on morphology and ITS sequence analysis. Additionally, its ability to dissolve Ca3(PO4)2 was assessed through NBRIP liquid culture, along with evaluations of nitrogen fixation, potassium solubilization, and IAA production. A pot experiment was conducted to determine the fungus's growth-promoting effects on barley. Key findings from the study included the successful isolation of a strain named W41, which exhibited strong phosphorus-solubilizing capabilities and was initially identified as Botryotinia ranunculi. In NBRIP liquid medium, the strain demonstrated an effective phosphorus content of 55.82 mg/L, along with nitrogen fixation, potassium solubilization, and IAA production abilities. The best germination and growth performance of barley seeds occurred when they were exposed to a spore liquid concentration of 1×103 cfu/mL in dish germination tests. In potting experiments, strain W41 significantly increased the average plant height, root length, fresh weight, and dry weight of barley seedlings by 29.71%, 28.69%, 48.49%, and 39.12%, respectively, compared to the control group. This research underscored the substantial phosphorus-solubilizing and growth-promoting potential of strain W41, offering a promising avenue for enhancing barley yields through the application of fungal agents.
