印度梨形孢培养基的优化
Optimization for Culture Medium of Piriformospora indica
目前培养印度梨形孢多采用马铃薯葡萄糖琼脂(PDA)培养基。为获得更适合印度梨形孢生长的培养基,研究固、液两种培养方式下,马铃薯、桃子、西瓜、草莓、苹果、玉米、红色圣女果和黄色圣女果8种天然有机添加物对印度梨形孢形态和生长速度的影响。结果表明,在添加200 g/L玉米的固体培养基中,印度梨形孢菌丝体生长最快;在玉米液体培养基中,菌丝体多呈大小比较一致、圆而紧密的菌丝球,培养至5 d,菌丝体鲜重、干重最高,分别为24.09 g、1.98 g;玉米是促进印度梨形孢生长的最佳天然有机添加物,马铃薯其次,PDA中培养至第5 d,菌丝体生物量达到最大,鲜重、干重分别为16.37 g、1.24 g。研究结果旨在为高效培养生产印度梨形孢提供重要理论依据。
At present, potato glucose agar medium is commonly used to culture Piriformospora indica. In order to explore a more suitable medium for the growth of P. indica, the effects of eight natural organic additives, including potato, peach, watermelon, strawberry, apple, corn, red cherry tomato, and yellow cherry tomato, on the morphology and growth rate of P. indica under two culture methods, solid and liquid, were studied respectively. The results were as follows: In the solid culture medium supplemented with 200 g/L corn, the mycelium of P. indica grew fastest. In corn liquid culture medium, the mycelium mostly appeared as relatively uniform, round and compact mycelium pellets. After cultured for 5 d, the fresh and dry weight of the mycelium were the highest, with 24.09 g and 1.98 g, respectively. Corn was the best natural organic additive to promote the growth of P. indica, followed by the potato, and after cultured for 5 d in PDA culture medium, the mycelium biomass reached its maximum, with the fresh and dry weight of 16.37 g and 1.24 g, respectively. The research results aim to provide an important theoretical basis for efficient culture and production of P. indica.
