西藏野生宽叶韭花粉离体萌发特性研究
A Study of the In Vitro Germination Characteristics of Wild Allium hookeri Pollen in Xizang
为明确野生宽叶韭花粉的离体萌发特性,以西藏宽叶韭花粉为研究对象,先使用单因素试验初筛花粉离体萌发培养需要的适宜光照条件、温度、时间以及培养基中蔗糖、硼酸、氯化钙的适宜质量浓度范围,再利用正交试验检验各成分之间的交互作用对花粉萌发的影响,筛选出花粉离体萌发的最适培养基。结果表明,25 ℃下暗培养1 h,更利于花粉萌发;单因素试验结果表明,适宜宽叶韭花粉萌发的培养基中蔗糖质量浓度为100 ~ 150 g/L,硼酸质量浓度为100 ~ 150 mg/L,氯化钙质量浓度为0 ~ 50 mg/L。试验中,蔗糖100 g/L、硼酸100 mg/L、氯化钙0 mg/L下花粉萌发率是最高,为57.39%,蔗糖100 g/L、硼酸200 mg/L、氯化钙150 mg/L下花粉萌发率是最低,为34.26%;蔗糖50 g/L、硼酸100 mg/L、氯化钙0 mg/L下的花粉管最长,达40.76 µm,蔗糖150 g/L、硼酸200 mg/L、氯化钙150 mg/L时的花粉管最短,仅19.11 µm。综上所述,宽叶韭花粉离体萌发最佳培养条件为25 ℃下暗培养1 h,最佳培养基组合为蔗糖100 g/L + 硼酸100 mg/L + 氯化钙0 mg/L。
To investigate the in vitro germination characteristics of wild Allium hookeri pollen, this study focused on Xizang A. hookeri pollen. First, single-factor experiments were conducted to preliminarily identify optimal light conditions, temperature, incubation time, and suitable concentration ranges of sucrose, boric acid, and calcium chloride in the culture medium for pollen germination. Subsequently, orthogonal experiments were employed to evaluate the interactive effects of these components on pollen germination and to screen for the optimal medium formulation. The results showed that dark conditions were more conducive to pollen germination, the optimal germination temperature was 25 ℃, and the optimal culture time was 1 h. The results of single factor test showed that sucrose concentration was 100 ~ 150 g/L, boric acid concentration was 100 ~ 150 mg/L and calcium chloride concentration was 0 ~ 50 mg/L in the suitable medium for A. hookeri pollen germination. In the orthogonal test, the highest pollen germination rate was 57.39% under sucrose 100 g/L, boric acid 100 mg/L and calcium chloride 0 mg/L, and the lowest pollen germination rate was 34.26% under sucrose 100 g/L, boric acid 200 mg/L and calcium chloride 150 mg/L. The pollen tube under the conditions of sucrose 50 g/L, boric acid 100 mg/L and calcium chloride 0 mg/L was the longest (40.76 μm), while the pollen tube under the conditions of sucrose 150 g/L, boric acid 200 mg/L and calcium chloride 150 mg/L was the shortest (19.11 μm). Platyphylla, while high concentration of calcium chloride could inhibit it. In conclusion, the optimal culture conditions for in vitro germination of A. hookeri pollen were dark culture at 25 ℃ for 1 h, and the optimal medium combination was sucrose 100 g/L + boric acid 100 mg/L + calcium chloride 0 mg/L.
