砂生槐实生无菌苗获得方法研究

砂生槐实生无菌苗获得方法研究
A Study of the Method of Obtaining Solid Sterile Seedlings of Sophora moorcroftiana
为建立砂生槐外植体消毒、组织培养快繁体系,本研究以砂生槐成熟种子为外植体,通过分析氯化汞(HgCl2)和次氯酸钠(NaClO)两种常用消毒剂在不同浓度不同时间对砂生槐种子外植体萌发指数、污染率的影响,得到砂生槐实生无菌苗的优化方案。结果显示,氯化汞(HgCl2)处理组的表现最优,发芽率为52.6%、发芽势9.20%、污染率为20%,但种苗长势一般;次氯酸钠(NaClO)处理组中污染率较高,但发芽率、发芽指数、发芽势均高于氯化汞处理组。结果表明,氯化汞(HgCl2)溶液对砂生槐种子的萌发具有抑制作用,次氯酸钠(NaClO)溶液的腐蚀作用则促进砂生槐种子的萌发。
To establish a disinfection and tissue culture rapid propagation system for explants of Sophora moorcroftiana, this study used mature seeds of S. moorcroftiana as explants. By analyzing the effects of two commonly used disinfectants, mercury chloride (HgCl2) and sodium hypochlorite (NaClO), at different concentrations and times on the germination index and contamination rate of S. moorcroftiana seed explants, an optimized scheme for S. moorcroftiana seedling without bacteria was obtained. The results showed that the mercury chloride (HgCl2) treatment group performed best, with a germination rate of 52.6%, a germination potential of 9.20%, and a contamination rate of 20%, but the growth of the seedlings was average. The contamination rate was relatively high in the sodium hypochlorite (NaClO) treatment group, but the germination rate, germination index, and germination potential were all higher than those in the mercury chloride treatment group. The results showed that the mercuric chloride (HgCl2) solution had an inhibitory effect on the germination of S. moorcroftiana seeds, while the corrosive effect of sodium hypochlorite (NaClO) solution promoted the germination of S. moorcroftiana seeds.
