四种提取青稞种子DNA方法的比较研究
Comparative Study of Four Methods for DNA Extraction from Highland Barley Seeds
为验证项目组自主研发的一种保持种子活力的玉米单粒胚乳DNA提取方法(简称“专利法”)在青稞种子中的适用性,本研究以不同青稞品种为材料,分别采用传统CTAB法、改良CTAB法、碱煮法及专利法提取种子DNA,并通过琼脂糖凝胶电泳和紫外分光光度法对所得DNA的浓度和纯度进行检测。同时,以提取的DNA为模板,进行普通PCR、多重PCR及实时荧光定量PCR检测,扩增了与淀粉合成、抗旱性和抗病性相关的5个基因。结果表明,利用专利法提取的青稞种子DNA浓度较高、纯度良好,在目标基因扩增中表现出优良的扩增效果,完全满足后续分子生物学实验的要求。同时,切除部分胚乳的种子仍可正常萌发与生长,可用于后续相关研究。本研究拓展了专利法在作物种类中的应用范围,并为青稞分子生物学研究提供了一种可保持种子活力的高质量单粒胚乳DNA提取方法。
To verify the applicability of a self-developed maize single-seed endosperm DNA extraction method that preserves seed viability (referred to as the "patented method") in barley seeds, this study used different barley varieties as materials. DNA was extracted from the seeds by the traditional CTAB method, the modified CTAB method, the alkaline boiling method, and the patented method, respectively. The concentration and purity of the obtained DNA were detected by agarose gel electrophoresis and ultraviolet spectrophotometry. Furthermore, wiht the extracted DNA as a template, conventional PCR, multiplex PCR, and real-time fluorescent quantitative PCR were performed to amplify five genes related to starch synthesis, drought resistance, and disease resistance. The results showed that the DNA extracted from barley seeds by the patented method had a high concentration and good purity and exhibited excellent amplification efficiency in the target gene amplifications, fully meeting the requirements for subsequent molecular biology experiments. Meanwhile, seeds that had undergone partial endosperm excision could still germinate and grow normally and could be used for subsequent related research. This study expands the application range of the patented method to other crop types and provides a high-quality single-seed endosperm DNA extraction method that preserves seed viability for molecular biology research in barley.
