机械门控离子通道Piezo1蛋白在缺血性心脏疾病中的研究进展
Research Progress of the Mechanosensitive Ion Channel Piezo1 in Ischemic Heart Disease
缺血性心脏病(IHD)是全球主要死亡原因之一,其发生发展与机械应激、钙稳态失衡、炎症反应及细胞死亡密切相关。Piezo1作为机械门控阳离子通道,在心肌细胞、内皮细胞、成纤维细胞和免疫细胞中广泛表达,可将机械刺激转化为Ca2+和活性氧信号,参与心肌梗死及缺血再灌注损伤中的炎症放大、线粒体功能障碍、纤维化和心室重构。本文综述Piezo1的结构、生理功能及其在IHD中的作用机制,总结了其靶向治疗的研究进展与现实障碍。
Ischemic heart disease (IHD) is one of the leading causes of death worldwide, and its onset and progression are closely associated with mechanical stress, calcium homeostasis imbalance, inflammatory responses, and cell death. As a mechanosensitive cation channel, Piezo1 is widely expressed in cardiomyocytes, endothelial cells, fibroblasts, and immune cells. It converts mechanical stimuli into Ca2+ and reactive oxygen species signals and participates in inflammatory amplification, mitochondrial dysfunction, fibrosis, and ventricular remodeling during myocardial infarction and myocardial ischemia-reperfusion injury. This article reviews the structure and physiological functions of Piezo1, as well as its mechanisms of action in IHD, and summarizes recent advances and current obstacles in Piezo1-targeted therapy.
